Actinomycetes from salt environment are the potential good material used for exploring new species and new product research, they can produce exocellular polysaccharide and other polymer, and protein content is extremely high, also have a lot of antibiotics, vitamin etc. Actinomycetes from salt environment will be screening new biological material, especially new antibiotics that were important resources. There are large areas of saline-alkali soil and salt lakes in China’s western region, Xinjiang and Qinghai provinces have many extreme high salt environment. The aim of This study is to investigat the diversity of actinobacteria in Balikun Lake and Qijiaojing lake. The diversity of actinobacteria in the soil from two lakes was investigated by culture-independent method based on phylogenetic analysis of16SrDNA gene sequences and selective isolation method. The study can be comprehensive excavating microbial species of salt lakes, and objective understanding to microbial species diversity and difference, also can accumulate useful species resources information, provide premise and security for the environment of salinization ecological restoration, extreme environment protection and utilization of resources.(1) The total DNA of two big salt lakes soil amplify the16SrDNA gene, and construct16SrDNA gene library. In the library insert fragments analysised by RFLP (Restriction Fragment Length Polymorphism, Restriction Fragment Length Polymorphism) and sequenced. The results show that:Balikun salt lake232clones, Qijiaojing salt lake176clones were actually the Actinobacteridae, Acidimicrobidae and Rubrobacteridae; Advantage actinomyces respectively are lamia of Iamiaceae, and Amycolatopsis; There were respectively81.0%,58.3%of clone sequences have published effective strains sequence comparability less than97%, most likely represents actinomyces new groups.(2)55strains were isolated by eight different selective medias. Balikun salt lake and Qijiaojing salt lake respectively obtain20,35strains actinomyces,20strains of Balikun salt lake belong to six species, respectively is Streptomyces, Nocardiopsis, Nesterenkonia, Actinopolyspora, Cellulomonas and Mycobacterium;35strains of Qijiaojing belong to seven species, the main are Streptomyces and Nocardiopsis, the others are Saccharomonospora, Saccharopolyspora, Kocuria, Brachybacterium and Actinopolyspora.(3)55strains actinomyces obtained from two salt lakes were though Polyketide synthase gene screening and detection, the results indicated that the positive rate was43.6%, among which Streptomyces and Nocardiopsis are the main sources.The results show that the two lakes between Tianshan mountain basin were has high diversity of Polyketide synthase gene, worthing further research and development.
