Rhamnolipid, which was produced by Pseudomonas aeruginosa, could not only be used in emulsifying, solubilizing, reducing surface activity, but have the ability of antibacterial and anti-tumor. But its low production and high cost limited the further research. So it becomes a effective way that screening high-yield rhamnolipid strains and detecting the activity component in the fermentation broth to reduce the cost. In this experiment, Strain M14808, which has a high rhamnolipids productivity, was screened from the mutants of P. Aeruginosa N1207after it being cultured in a Superconducting Magnet. The fermentation of the strains was collected and isolated, and the activity of each components were detected. The result of this research is as follows:1.Laboratory preserved strains N1207was identified by microbial morphology, physiology and biochemistry,16S rDNA. The bacterial was gram-negative rod-shaped; Evolutionary relationship analysis showed that the closest homologous bacteria was P. Aeruginosa S164S relationship. According to the result the bacterial was named P. Aeruginosa N1207.2. A high product of Rhamnolipid strain M14808was screened after P. aeruginosa N1207was mutagenized in the Superconducting Magnet; and then fermented in a optimal conditions. The fermentation was collected and isolated. Five component was got, they were moni-and di-rhamnolipid, yellow and blue pigment, and lipopeptides.The biological activity of the five kinds of components were detected. Antimicrobial Experimental results showed, the lipopeptide had a strong ability to inhibit Gram-negative, Gram-positive bacterial and fungi, the MIC was6μg/ml. The antitumor experimental results showed that the blue pigment is acting a lowest IC50values15.67±0.13μg/ml on cell line HepG2. Lipopeptide role in cell A549IC50was82.039±2.06μg/ml, lower than the other three component.3.The two component were characterized by MS or FTIR, it showed the blue pigment was pyocyanin. The lipopeptide has carbon chain and amide bond. The two components could promote cancer cells A549and HepG2apoptosis respectively.
