Lipase is widely used in biocatalysis. It is widespread among animals, plants and microorganisms. The most widely used lipases come from microorganisms.Based on the mature peptide sequence of lipase from Proteus sp. and the preference of Pichia pastoris to codons, the full oligonucleotieds sequence coding the lipase from Proteus sp. were designed and synthesized. The synthesized gene, named as SW1, was proved to be the same as the designed by sequencing. Then the SW1gene was cloned into the expression vector pPIC9K.The recombinant plasmid pPIC9K-SWl was linearized with restriction enzyme Bgl II and transformed into Pichia pastoris strain GS115. The stably-transformed Pichia pastoris strain GS115was verified by PCR and substrate screen. Furthermore, the recombinant lipase was verified to be successfully induced to secretly express in Pichia pastoris strain GS115by SDS-PAGE analysis.
