7-aminocephalosporanic acid (7-ACA) is important nucleus for synthetizingcephalosporins antibiotic. Cephalosporin C acylase (CCA) is key enzyme to manufacture7-ACA only through one step. It can directly catalyze raw material cephalosporin togenerate7-ACA.Researches about production process of CCA have important practical significance toproduce7-ACA through one step. This paper is mainly carried out a study of the processof separation&purification and immobilized of cephalosporin C acylase.First, chemical permeation method was determined the optimum extraction process andthe next best method was homogenization. Single factor test showed that the most suitableamount of penetrant was1.0%and the most suitable osmosis time and temperature were60minutes and35℃respectively. And the best homogenization pressure was determinedto850bar. Moreover,45℃and60minutes are confirmed as the best temperature andduration for heating treating. The concentration of ammonium sulfate is50%.Then,research about the main factors effecting homogenization process were taken.Comparing four different types of carrier, HC-01and ES-2were chosen. The optimumconditions through orthogonal experiments were determined as: immobilizationtemperature24℃~26℃; phosphate concentration1.0M; enzyme ration1000:1.Further, reaction condition to manufacture7-ACA through one step CPC enzyme wasdetermined to: pyrolysis temperature20℃; pH8.3; reaction time about90min; the aboveresearch lay the foundation for the one step enzymatic production of7-ACA.
