Oxalic acid oxalate decarboxylase can catalyze the decomposition of formic acid and carbon dioxide, the application of the enzyme concentrated in the papermaking wastewater oxalate, degradation of oxalic acid, bio-detection and so on. However, the current production of oxalate decarboxylase limits the application of the enzyme. In order to understand the enzyme induction mechanism, enhance the enzyme production; this paper studied the acid on Tramete versicolor induce conditions, and enzyme extraction conditions were optimized, the stability of the enzyme was measured. Bioreactor has been used to study the enzyme induce conditions. Mycelium tries to use induced by phytic acid and straw hydrolyzate has been used as a carbon source to improve enzyme production in cultured mycelium.At first, Tramete versicolor induction conditions were optimized, the main research on the yield of oxalic acid concentration, and then add the use of oxalic acid and fed in batches to improve the enzyme production. By measuring the cell growth process in the different parameters (for example, enzyme activity, protein content, biomass, etc.) to compare the different methods of inducing enzyme production under the conditions of advantages and disadvantages, of which 10 mM activity in the highest,12.6 U/L fermentation broth,20 mM protein, the highest specific activity for 27.54 U/mg; and batches to add oxalic acid, enzyme activity up to 1086 U/L fermentation broth, protein specific activity reached 65.6 U/mg, after feeding up to 2060 U/L fermentation broth, protein specific activity reached 75.17 U/mg. Bioreactor culture Tramete versicolor, ventilation volume 4 L/min, pH 5.0, for better culture conditions, oxalic acid induced broth pH 3.5 on the production of beneficial enzymes. With oxalic acid as an acid pH value of fermentation broth adjusted to induce enzyme production can maintain a more stable enzyme production.At last, Tramete versicolor oxalate decarboxylase fragmentation conditions and the extraction conditions were optimized, using ultrasonic fragmentation method in order to work/ rest ratio,6 s/4 s better, you can reach liquid nitrogen grinding 98% of the crushing efficiency, using Tween -80 buffer solution can be slightly increased rate of extraction of the mycelia debris, but after dialysis losses. Use of phytic acid on Tramete versicolor for induction, can produce a small amount of oxalic acid decarboxylase. The use of straw hydrolyzate cultivation can greatly enhance the mycelial biomass,10 mM oxalic acid concentrations induced activity reached 39.2 U/L fermentation broth, protein specific activity of 12.14 U/mg.
