Semen Zizyphi Spinosae is the dried ripe seed of ziziphus jujuba in Rhamnaceae plant,which exhibits sedative,hypnotic,peaceful activity.Total saponins of Semen Zizyphi Spinosae are important with respect to pharmacological effect of Semen Zizyphi Spinosae.Jujubosides A is the main components in Semen Zizyphi Spinosae to the sedative activity.The main results of these studies are as follows:1. The spectrophotometry method is accurate and convenient.The Ultraviolet absorption of Saponin is weak,vanillin acetum perchloric acid system is the common chromogenic reagent.After chromogening the max absorption wavelength would be determined in visible region,combining with PLS which shows the extremely ability on extract of information.In the average recoveries of jujubosides A and B in the simulated samples were 85.54 % and 87.26 %, and the RSDs were 4.46 % and 3.47 %. It can be applied to determine jujubosides A and B in zizyphi spinosi semen.2. Molecular imprinting technique is simulate recognition between the natural enzyme and substrate.To the target as a template,By way of non-covalent binding of monomers and templates,add crosslinker and initiator prepared molecularly imprinted polymer,Eluting after the end of the template molecule reaction can form a hole which is capable of identifying the molecular template imprinted polymer.Molecularly imprinted membranes (MIMs) provide a feasible and effective approach of specific target molecule,developing jujubosides A molecularly imprinted composite membranes using surface modified polymerization by UV irradiation in the presence of jujubosides A as template,followed by a deposition of molecularly imprinied polymer layer on the surface of polyvinylidene fluoride microfiltration membranes.Molecular imprinted polymers were synthesized with Jujubosides A as template molecule,Acetonitrile as solvent,MAA as functional monomers,EGDMA as the crosslinker,AIBN as initiator. Optimized reaction conditions:the optimum molar ratio of template,functional monomer(MAA) and crosslinker(EDMA) is 1:28:140;Reaction temperature is 60 degrees;Reaction time is 48 hours.In this condition,the QMAX of synthesized molecularly imprinted membranes for jujubosides A could achieve 13.2μmol/cm~2.3. Jujubosides A-molecularly imprinted membranes work in two ways which including interception way and penetration way. In the infiltration way, The transit dose and speed which molecularly imprinted composite membrane for jujubosides A were greater far and away than non-imprinted composite composite membrane. The arrange of functional group sincavity was disorganized,the combining eapacity of non-imprinted composite membranes for jujubosides A was lower than moleculariy imprinied composite membranes.In the penetration way, under the optimized conditions, could get the jujubosides A of 91.9% purity. In the infiltration way, could get the jujubosides A of 88.7% purity, jujubosides B could not through the membranes.This method more suitable for a large amount of purified workplace.
