It includes the following three chapters:In the first chapter, recent development of the design, synthesis and biological activitystudies of novel nucleosides, especially5-substituted pyrimidine nucleosides and carbocyclicnucleosides, were briefly reviewed.In the second chapter, a series of2’-deoxy-5-substituted carbocyclic uridine derivativeswere designed and synthesized. The synthesis of these compounds includes the followingsteps:1) Synthesis of cyclopentenol from cyclopenta-1,3-diene;2) synthesis of N3-protectedthymine;3) Mitsunobu coupling of cyclopentenol with N3-protected thymine;4) synthesis of5-formyl-2’-deoxy carbocyclic uridine;5) synthesis of the target compounds.In the third chapter, a series of2’,3’-dideoxy-5-substituted carbocyclic uridine derivativeswere designed and synthesized.
