Studies On The Chemical Constituents And Quality Standards Form The Roots Of Rosa Laevigata Michx

Traditional Chinese medicine Rose laevigata Michx. is the root orroot skin of Rose laevigata Michx in Rosaceae. Root of the Roselaevigata Michx being used as medicine is of a long history. It wasrecorded in《The Chinese herbal》,《Compendium of Materia Medica》and so on. And it is one of Guangxi Chinese herbal medicine in largeamount. In this paper, the chemical constituents in roots of the Roselaevigata Michx and their high contents of an isomers have beenresearched. Also, its quality standards have been studied primarily. Themain points of the three parts are described as follows.1. In this paper, the chemical constituents of the70%ethanol extractswere separated and structural identified. By the means of the columnchromatographic polyamide, silica gel, ODS, Sephadex LH-20gelfiltering and other separation means,12compounds were isolated. At thesame time, with heavy crystallization, TLC and HPLC to aididentification sample purity and through the ESI-MS,1H-NMR,13C-NMR spectrum and chemical structure analysis,5of12compoundswere structural identified as follows:2α,3α,19α-2,3,19-tridroxyurs-12-en-28-oic acid β-D-glucopyranosylester,2α,3β,19α-2,3,19-tridroxyurs-12-en-28-oic acid β-D-glucopyranosyl ester，2α,3β,19α-trihydroxyurs-12-en-28-oic acid β-Dglucopyranosylester, ursolic acid, gallic acid.2. In Roots of the Rose laevigata Michx. the contents of2α,3α,19α-2,3,19-tridroxyurs-12-en-28-oic acid β-D-glucopyranosylester(saponins D) and2α,3β,19α-2,3,19-tridroxyurs-12-en-28-oic acidβ-D-glucopyranosyl ester(saponins E) are more than the others. So theextraction technology conditions optimization of the isomers has beeninvestigated by HPLC method. And the results show that: the optimumextraction conditions are the mass fraction of with75times amount of70%ethanol, the ultrasonic extraction90minutes, saponins D content0.7236%on average, saponins E content0.8871%on average.3. Roots of the Rose laevigata Michx. quality standards has beenresearched. The qualitative and quantitative research on JinYing rootform11origin of the materials had been made. And through the researchthe quality control standards has been established.
