In this paper, studies on preparation and biological activity of hydrolyzed protein ofPatinopecten yessonensis skirt, and preliminary separation of the product by use of gelchromatography was carried out, which will provide reasonable and scientific basis for thedeveloping and utilizing of Patinopecten yessonensis skirt. The results showed as follows:1. The content of total protein of Patinopecten yessonensis skirt was11.23%, moisture was85.67%, ash was0.96%.2. In the experiment, neutral proteinase, papain, acid protease and trysin were used tohydrolyze Patinopecten yessonensis skirt. The optimal proteases were neutral proteinase andtrysin.3. Orthogonal experiment was employed to optimize single enzyme hydrolysis condition ofPatinopecten yessonensis skirt. The effect of hydrolysis temperature, hydrolysis time, enzymedosage, pH and liquid/solid ratio on hydrolysis was studied. The result indicated that the optimaltechnological condition of neutral proteinase was: hydrolysis at temperature45℃, neutralproteinase dosage E/S=1.1%, pH8.0, hydrolysis time6h, liquid and solid ratio3:1, the degree ofhydrolysis (DH) was34.58%under the condition; the optimal technological condition of trysinwas: hydrolysis at temperature50℃, trysin dosage E/S=1.0%, pH8.0, hydrolysis time6h, liquidand solid ratio3:1, the DH was28.70%under the condition.4. Orthogonal experiment was employed to optimize compound enzymes hydrolysisconditions of Patinopecten yessonensis skirt of the two enzymes choosen in the experiment. Theliquid and solid ratio was fixed as3:1, and the effect of hydrolysis temperature, the total enzymedosage, the proportion of enzymes, pH, hydrolysis time was studied. The optimal condition ofcompound enzymes was: hydrolysis at temperature45℃, the quantity of enzymes addedE/S=1.1%, the proportion of neutral proteinase and trysin2:1, pH7.5, hydrolysis time6h, liquidand solid ratio was3:1, the DH was37.69%under the condition. The results revealed thatcompound enzymatic hydrolysis was better than single enzymatic hydrolysis.5. The antioxidant activity and antifungal activity were evaluated, and the results showedthat the hydrolyzed protein of Patinopecten yessonensis skirt had significant hydroxyl radicalscavenging activity, with a half-scavenging concentration (EC50) of0.2981mg/mL; and it hadcertain antifungal activity against Stemphylium solani weber and Phomopsis asparagi.6. The hydrolyzed protein of Patinopecten yessonensis skirt was separated and purifiedprimarily, and Sephadex G-25column was used to analyze the distribution of peptides which had antioxidant activity, the results confirmed that at least three kinds of antioxidant peptides ofdifferent molecular weight exist in the enzymatic solution.
