The immunoassay technology has been widely used in various research fields, due toits many features such as no radiation, simple operation, high sensitivity and so on. Inthis paper, four kinds of drugs, lidocaine, metronidazole, lamivudine, zidovudine, astarget analytes, are labeled with fluorescent reagent. And then, immunofluorescenceassay was established on the basis of competitive adsorption of the drug ant itsfluorescent derivative against corresponding molecularly imprinted polymer. Theresearch work consists of several parts as following:1. The common analytical methods, such as HPLC, of drugs mentioned abovewere briefly reviewed. And the development of the immunoassay technologiesespecially immunofluorescence analysis technique which are widely used in druganalysis, as well as fluorescent labeling technology were introduced.2. Fluorescent analogues were prepared agaist lidocaine which has not activegroups in the molecular for ready derivatiazation for fluorescence labelling. Theexperimental results showed that the synthesized fluorescent analogs showed goodfluorescence performance.3. Using simple aromatic compounds as raw materials,9-aminoacridine wassynthesized by the feasible reaction scheme. And the UV absorption spectrum andfluorescent spectrum of the compound were recorded. The result indicated that9-aminoacridine showed the good fluorescence performance, and it can be used as anideal fluorescent labeling reagent for the drugs.4. In order to minimize the affect of labeling reagent on the biding sites of thedrug molecules against the imprinted polymer, and to decrease affect of the drugmolecule on the fluorescence properties of the labeling reagent, two strong reactivecompounds were selected as a spacer. According to their reaction mechanisms,synthetic route was designed for the preparation of three fluorescent derivatives, andthe fluorescent labeled drugs were successfully synthesized. To some reaction with low yield, a series of parallel experiments were designed to select optimal conditions,and the reaction yield was increased.5. The fluorescence properties of prepared three fluorescent markers such asmetronidazole were investigated. The results showed that the compounds displayedgood fluorescence properties, a standard curve was produced in acceptableconcentration ranges. And the static adsorption experiment was carried on drugs andfluorescent markers to the corresponding polymer. On the basis of the experimentaldata, metronidazole and zidovudine fluorescent markers as competitive reagents, therespective molecularly imprinted polymers as "antibodies", the competitiveadsorption experiments was completed. The results showed that the relationshipbetween the concentration of the drugs and the relative fluorescence intensity possessgood linear relations.
