Crataegus oxyacantha, also known as English hawthorn is part of the Rosaceae plant family. Works including the extraction oftotal triterpenoids derived from hawthorn have been reported in this paper. On this study, the raw materials were hawthorn, with different methods of extraction, separation, purification and component analysis. The research lays foundation for development and utilization of hawthorn, the major results as follows:1. Reflux extraction was applied to extract total triterpenoids from hawthorn. The optimal extraction:ethanol concentration of85%, extraction time2h, extraction temperature85℃, and solid-liquid ratio of7mL·g-1. The triterpenoids yield under the optimum conditions was found to2.57±0.4%.2. Supercritical fluid extraction was applied to extract total triterpenoids from hawthorn. The optimal extraction:extraction pressure30MPa, extraction temperature45℃, extraction time1.5h, modifier carrier15mL, under these conditions, with yield rate0.97%.3. Microwave-assisted extraction was applied to extract total triterpenoids from hawthorn, the optimal extraction:methanol concentration of60%, microwave power640W, solid-liquid ratio of17mL·g-1, microwave time of97s, with yield rate4.08%.4. Comparison of different methods for extraction of total triterpenoids, which showed supercritical-reflux-microwave extraction was the best extraction method, with yield rate5.13%.5. Component analysis:The supercritical fluid extract were analyzed by means of GC-MS,88volatile compounds were identified from hawthorn, among them,66kinds of components for the first time detected in hawthorn.The hawthorn extract were analyzed by means of GC-MS,50compounds were identified from hawthorn, among them,39kinds of components for the first time detected in hawthorn.The compounds in crude extract hawthorn were separated and purified by silica gel column and Sephadex LH-20, and analysised by IR、LC-MS、1H NMR and13C NMR spectrometric techniques, compositions in crude extract were elucidated as follows:triterpenoids, flavonoids, ketones, steroidal hydrocarbons, aldehydes, alcohols, esters, and acids were the main compounds observed in hawthorn. Ursolic acid and squalene are triterpenoids, and squalene was determined for the first time.
