Feruloylated oligosaccharids (FOS) is the compound that link with ferulic acid andoligosaccharides through the ester bond, so FOShave the physiological function of the twosubstances. FOSis water-soluble, with non-ion attribute properties, strong permeabilitybecause of the special ester linkage,so they can enter into human body mitochondria to playantioxidant effect easily. Corn bran is used as the raw material in this paper, the extraction,isolation, purification, and the antioxidant activities of FOSfrom corn bran were discussed.The main research contents and conclusions are as follows:(1)The production technology of composite cellulose enzymic preparation of corn branFOSwas reseached. There were five factors as the enzymic system pH value, temperature,enzyme quantity, substrate concentration and reaction time.In response to the FOSconcentration, the conditions of the extraction were optimized by the response surfaceanalysis of five factors four level on the base of single factor experiment and the best reactionconditions for composite cellulose enzymic preparation of corn bran were pH5.30, reactiontemperature51℃, enzyme concentration7.62%, substrate concentration11.03%, reactiontime42h and under the conditions, the content of the FOSwas23.934u mol/g.(2) The production technology of microwave radiation pretreatment of corn bran toprepare FOSwas reseached. There were five factors as the microwave pretreatment pH value,radiation time, radiation power and liquid material. In response to the FOSconcentration, theconditions of the microwave radiation pretreatment were optimized by the response surfaceanalysis of four factors three levels on the base of single factor experiment and the bestreaction conditions for microwave radiation pretreatment were pH0.6, microwave radiationpower622.4W, liquid than material10:1, microwave radiation time8min. Corn bran wasenzymic hydrolysis after microwave radiation under the best enzymolysis conditions, thecontent of the FOSwas42.160u mol/g.Then the production technology of production of cornbran FOSby microwave-assisted enzymic hydrolysis was reseach. There were five factors asthe enzymic system pH value, temperature, and enzyme quantity, substrate concentration,reaction time. In response to the FOSconcentration, the conditions of the extraction wereoptimized by the response surface analysis of four factors three levels on the base of singlefactor experiment and the best reaction conditions for production of corn bran FOSbymicrowave-assisted enzymic hydrolysis are the enzyme solution pH5.0, temperature50℃,enzyme concentration5.64%, substrate concentration8.05%, enzyme time36h. Under theconditions, the content of the FOSwas up to119.509u mol/g.(3) The conditions of purification of corn bran FOSwere optimized by AmberliteXAD-2resin through static experiment and dynamic test and the best conditions were sample pH value5.0,the biggest resin adsorption quantity5.88u mol/mL, sample on flow velocity1mL/min, sample quantity86.78u mol, water-elution volume800mL,50%ethanol, Elutionvelocity8mL/min, elution time135min. Under the conditions, FOSrecovery rate reached91.64%.(4) The preliminary purification samples was further purified by Sephadex LH-20filtering gel chromatography. The conditions of purification are optimized through the singlefactor experiment and determined the best conditions for: elution agents25%ethanol, samplequantity50mg/mL1.5mL, flow rate0.5mL/min. Under the conditions, five componentswhich are identified FOSby HPLC and infrared spectrum analysis method are separated.(5) The antioxidant activity of corn bran FOSwas studyed. Include OH·scavengingactivity, O2-· scavenging activity, DPPH· scavenging activity, reductive ability, antioxidationactivity on oil.50%OH·scavenging activity was3.082mg/mL、50%O2-· scavenging activitywas mg/mL、50%DPPH· scavenging activity was0.7354mg/mL。...
