With the rapid development of industrialization of prebiotic products and national improvement of living standards, fermented dairy products plays an important role in people’s daily lives more and more. national demand for fermented dairy products, and consumption of demand prompted us to produce independentdirect inoculation of property rights to produce starter, which is more convenient, continuous, stable.By improving the solids concentration in the fermentation broth,to provide adequate nutrition to the bacteria, on the basis of quality scores fermentation broth for downstream processing,this will ensure a high number of viable cells,to avoid the probability of the second to add a protective agent to produce contamination,greatly improves production efficiency and reduce the cost of the output.This study is based on this reality,to optimize the medium composition and concentration in the fermentation process,control the fermentation conditions, operational processes of the simulation of industrial production,L. acidophilus and other probiotics will help to achieve production in the industrialized.Bring economic benefits to the enterprise and has important practical significance.has important practical significance.This study contains L. acidophilus basic physiological activity determination.culture medium research,filter of skimmed milk fermentation medium,centrifugal collection,and spray drying protection formula, we also examined the fermentation activity of the starter.The results indicated:simulating the human gastrointestinal tract, bile and acid resistance of L. acidophilus.The result showed that L. acidophilus has strong acid and bile resistant capability, can pass through gastro-intestine in high activity to play a probity role.Compare with industrial production medium on the growth of Lactobacillus acidophilus, Final choice of skim milk fermentation medium, determination of the optimum concentration of skim milk,based on the basis medium,optimized carbon source, and probiotics factor, got the best culture medium formula:25%powder of skimmed milk,2%Glucose,0.5%Oligo-ismaltose,adding distilledwater to1000mL.L.acidophilus could be cultivated to1.51×109cfu/mL cfu/mL, equivalent to MRS. With the high number of viable cells of strains of commercial development.Simulated industrial fermentation in laboratory Swiss3.7L fermentor,got the optimum fermentation conditions,temperature37.30℃,pH5.81,rotate speed120r/min.Inoculated3% L.acidophilus KLDSAD3-SU into culture,After three repeated experiments under this condition,the number of viable cells reach3.12x109cfu/mL,representing an optimization increased by36.19%.Prove accurate and reliable response surface method for optimization of fermentation conditions has practical value.Based on25%skim milk fermentation medium,spray drying protection of L. acidophilus has been studied,the optimum formula was determined as:Trehalose6%, Gelatin1%,glycerin2%,use of low-temperature spray drying tower spray drying, the inlet air temperature was110℃,the outlet air temperature was53℃.Number of viable cells can reach5.37×109cfu/g.By culture starter qualify test,we could find that:the viable count of spray drying starter during storage period remained stable.In the first4hours, spray drying starter don’t show rapid acid decrease as the liquid starter in the mink.But in the next few hours it showed rapid acid decrease.Sensory evaluation of L. acidophilus fermented milk.The overall evaluation of fermented milk organizations even delicate,silky,compared with sticky,slightly less acidity,lack of flavor of fermented milk.No precipitation, suitable for edible whey.
