Sonchus oleraceus L contains natural falconoids, which have the ability to lower blood pressure, to enhance immune regulation, anti-age, antioxidant. The Sonchus oleraceus L as raw material, the extraction of falconoids volume as the index, respectively studing the extraction conditions of falconoids through the ultra high pressure method, ultrasonic method, ethanol reflux; using macroporous resin to purify the total falconoids of Sonchus oleraceus L; respectively using DPPH system of free radical, superoxide anion, hydroxyl free radical system, system ABTS free radical system to evaluate antioxidant activity of Sonchus oleraceus L falconoids and its free radical scavenging ability; the main conclusions are summarized as follows:(l)Using ultra high pressure method, ultrasonic method, ethanol extraction to extract falconoids from Sonchus oleraceus L and the use of response surface method to optimize the extraction process respectively to determine the optimal extraction method, the experimental results show that:the ultrahigh pressure extraction not only the extraction time is the shortest, and extracting a quantity highest, up to5.753mg/g, the the optimum extraction process is:ethanol volume fraction of50%, holding pressure time8.32min, extraction pressure is446MPa, material to liquid ratio1:70(g/mL).(2)Using macroporous resin to purify Sonchus oleraceus L falconoids, through screening of eight kinds of macroporous resin in order to determine the most suitable purified resin of Sonchus oleraceus L falconoids, using response surface method to separately optimize the process of adsorption and desorption, and to determine the optimal purification process, the experimental results show that:the AB-8type macroporous resin is most suitable for purification of Sonchus oleraceus L falconoids; the optimum conditions:sample concentration3.73mg/mL, sample liquid rate2.31mL/min, sample liquid pH4.18, rate of adsorption is up to84.32%; the optimum conditions are:ethanol concentration of79.96%, desorption velocityl.25mL/min, ethanol elution volume6.38BV(column volume), the elution rate up to91.73%. Purity from34.78%to82.91%purity increased2.39times.(3)The use of DPPH,Superoxide anion,Hydroxyl ABTS free radical evaluation method to evaluate antioxidant activity of each part of Sonchus oleraceus L (root, stem, leaf) falconoids. The experimental results show that:each part of Sonchus oleraceus L (root, stem, leaf) falconoids have antioxidant activity, to compare antioxidant ability with the Vc,BHT positive control phase, on the ability to scaveng hydroxy free radical, stronger than the positive control, in scavenging the DPPH, ABTS free radical, superoxide anion free radical, Sonchus oleraceus L is same level with the positive control. Sonchus oleraceus L falconoids extracts is a good source of antioxidants.
