Soybean blight, also known as Phytophthora root rot, caused by Phytophthora sojae. It was soil-borne diseases which was.widely distributed, and extremely serious harm over the world. Phytophthora root rot was still an important disease that impact soybean production. Our laboratory found the secondary metabolites-borrelidin, produced by actinomycetes neau-D50can effectively inhibit the growth of Phytophthora sojae and identified its threonyl-tRNA synthetase (ThrRS) was the target of borrelidin. Accordingly, this article was committed to find the drugs that the target was the same to borrelidin but the dose was smaller, weaker side effects than it, we hoped it could apply in practical and developed the drugs which was more available and more scientific value for Phytophthora sojae,. The main findings as follows:1) Our test used CA medium (carrot medium), the result of the half inhibitory concentration (EC50) of borrelidin for Phytophthora sojae is0.0056μg/mL, and found it is able to completely inhibit the growth of bacterial when the concentration of borrelidin is0.02μg/mL. Compared with the EC50values of the Phytophthora infestans and E.coli DH5a which kept in our laboratory, the Phytophthora sojae was sensitive strain that resistant borrelidin.2) Added threonine to the CA medium containing certain concentration borrelidin, then found threonine could improve the growth rate of Phytophthora sojae that was inhibited by borrelidin from32.72%to67.19%, according to the experimental results and references, we could deduce the ThrRS of Phytophthora sojae was an important target of borrelidin.3) Accorded to the known ThrRS sequences of Phytophthora sojae to design primers, compared with the cloned gene sequences and known genes, the sequence is identical. Thus we determine the accuracy of ThrRS sequences of Phytophthora sojae. Then based on the sequence, we predicted the three-dimensional structure of ThrRS of Phytophthora sojae.4) Used SWISS-MODEL software for online prediction and received the three-dimensional structure of ThrRS of Phytophthora sojae.5) we used the Discovery Studio2.5(DS2.5) software carried out drug screening in the DrugBank database. We obtain50drugs that the fraction were greater than125, and chose the top five compounds as the target compounds of our screening at last.
