Biosurfactant is a kind of compound with surface active substance produced bymicroorganisms(bacteria, fungi and yeasts). In comparison with chemical synthesis surfactant,it has a lot of advantages,such as non-toxic or low toxic, environmental compatibility,biodegradable, structural diversity, stable under extreme environment.However, the fact thatthe costs of biosurfactant production is expensive and the yield is low have already becamethe bottleneck of large-scale production.So how to reduce the costs of biosurfactantproduction and improve its yield is a problem need to solve aurgently.This paper target abiosurfactant produced bacteria-Pseudomonas aeruginosa MZ01which was screened byresearch team as the study object,to optimize the conditions of biosurfactants production,explore the method to improve biosurfactant production by Pseudomonas aeruginosa MZ01and provide a reference method for large-scale production of biosurfactant. The main resultsare as follows:⑴Production of biosurfactant by Pseudomonas aeruginosa MZ01using familiarvegetable oil and agricultural wastes. The results show that the best concentrations offragrant sesame oil and nitrogen source(nitrates) were2%and0.5%,respectively.The mediumsurface tension was reduced to25.9mN/m in the best of concentrations carbon source andnitrogen.The study of production of biosurfactant by Pseudomonas aeruginosa MZ01fromseed oil cake shown, after10days cultivation,the surface tension of cell free broth (calciumfree) were32.7and33.4mN/m respectively,which indicates that rapeseed and flax seed oilcake are suitable for production of biosurfactant,but it need a long fermentation time and thesurface tension is higher.⑵The time course of Pseudomonas aeruginosa MZ01growth and biosurfactantproduction using fragrant sesame oil shown that the biosurfactant was the secondarymetabolite.The production of biosurfactant at the end of fermentation was newly84.5timesof the first day and the best pH is7.4-7.6.The biosurfactant producted by Pseudomonasaeruginosa MZ01using fragrant sesame oil was stability in extreme environment(hightemperature and high salinity), and extreme environment was beneficial to improving surfaceactivity.⑶Production of biosurfactant by Pseudomonas aeruginosa MZ01using corn oil in asequencing batch reactor.The surface tension of medium was reduced to27-30mN/m.When the cycle time was two days and the dosage of corn oil was20ml per cycle,cell growth andproduction of biosurfactant shown a growth–associated,and after ten cycle of operation, themaximum production of biosurfactant and biomass were10.6g/l and2.733(OD610),respectively.
