In recent years, with the development of biological molecules, the detection of biological molecules using DNA bio-sensors had been the hot spots in the life sciences and materials science. In the article, a highly sensitive method which combined nanotechnology and DNA bio-sensors was introduced. Due to the high sensitivity, easiness of fabrication and low cost, the method may provide an alternative tool for detection protein in clinical diagnosis.The main jobs of this paper can be concluded as follows:1. The synthesis and scale-up production of silver nanocubes and gold nanocagesIn this paper, two methods including ethylene glycol reduction of AgNO3and ethylene glycol reduction of CF3COOAg were used to synthesize silver nanocubes. The size of synthesized silver nanocubes was50-70nm.Compared two methods found that the latter could large-scale synthesis. Then, Au nanocages were prepared using a remarkably simple galvanic replacement reaction between HAuCl4and Ag nanocubes. Comparing LSPR peak and electron microscopy images, we obtained the optimum reaction conditions. The size of synthesized gold nanocubes was60-70nm. Finally, we achieved to scale-up synthesis on this basis.2. The fabrication of ATP biosensor based on the gold nanocages amplification technology and its application for ATP detection in cancer cellsIn this paper, the guest molecules were loaded in the hollow interiors of gold nanocages. To block the fluorescent molecules in the cages, the gold nanocages were wrapped with DNAs. Under the optimal conditions:the optimal temperature is37℃and the optimal reaction time is30min. The biosensor was able to detect ATP in the range of1.0×10-7to1.0×10-5M. The biosensor has great potential in the biomedical field and can be used for specific detection of different target molecules. It provides new ways and methods for early diagnosis and treatment of major diseases such as cancer.3. Fluorescence imaging by ATP probe in cellsThe guest molecules were loaded in the hollow interiors of gold nanocages. Adding the probe into tumor cells cultures, the Laser Confocal Scanning Microscope (LCSM) images of tumor cells were obtained. The images illustrate the release of fluorescent molecular in tumor cells. The system has the potential to apply to deliver drug and realize the controlled release of drug.
