The mainly problems existing in the ophthalmic drug delivery system is in the dose lost of drug, the short retention time and the low bioavailability. At present, the research trend of the ophthalmic drug delivery system is mainly through the improvement of biopharmaceutical drugs characteristics, increase the corneal contact time of drug and improve the bioavailability. This paper prepared different types of matrine phytosomes in situ gel and hope to improve regular ophthalmic preparation deficiency.In the matrine phytosomes study, screening and determined the matrine phytosomes preparation prescription:drug concentration20mg/ml, matrine:soy lecithin1:1, reaction temperature55℃; The physical and chemical properties of preliminary exploration proved, the lipid solubility. of matrine increased nearly three times, and significantly improve the drug absorption in the cornea.We optimized the prescription of different types in situ gel, the temperature sensitive in situ gel prescription:P40718%,P1887%, PEG60001%,mannitol5%,BKC0.02%and add pure water to the full amount; the ion sensitive in situ gel prescription:gellan gum0.2%, sodium alginate0.6%, mannitol5%, BKC0.02%, and add pure water to the full amount; pH sensitive in situ gel prescription:carbopol0.1%, HPMC0.8%, mannitol5%, BKC0.02%, and add pure water to the full amount.In the preliminary research of the in situ gel rheological properties we proved that the three types in situ gel are all pseudoplastic fluid, and suitable for the ophthalmic drug delivery system.We determine the in vitro release degrees and the permeability of in vitro rabbit corneal, the drug release speed of in situ gel is fast and the permeability is significantly improved; in the irritant and safety evaluation we proved irritant of the in situ gel had no irritant and good safety; at last we choose the temperature sensitive in situ gel as the ophthalmic drug delivery system, established quality standards and preliminary studied the stability of the temperature sensitive in situ gel.This research use phytosomes and in situ gel to improve the drug release performance of matrine, provide reference for matrine in the ophthalmology clinical application; systematic flitered and comprehensive compared the matrine in situ drug delivery system,filled the blank of the related research.
