Aralia elata (Aralia etata (Miq) Seem),also known as Liaodong Aralia,commonly known as "the barbed old Shoots",belong to Araliaceae,is a kind of local herbs that can be easily found in Jilin Province, having the effect of invigorating qi for tranquilization,striving sperm and nourishing kidney,dispelling wind and eliminating dampness,promoting blood circulation to arrest pain,invigorating stomach and profiting water;The tender shoot in spring can serve as a food delicacies,with strong health caring function.Aralia elata Monomer saponins acid-3-O-beta-D-pyran glucuronide (CE) were isolated from Aralia elata total saponins combined with macroporous adsorptive resins and chemical conversion methods.Our laboratory has already demonstrated that CE plays role of treating arrhythmia.In this study we used the Langendorff isolated heart perfusion techniques in order to study effect of CE on isolated heart different models of rats, as an effective drug that may have an anti-arrhythmic effect.The study were divided into two parts:(1)Effectt of CE on the hemodynamics of normal isolated rat heart modelWe took CE as samples,and built the Langendorff model with isolated rats hearts and observed the hemodynamic parameters of each group:left ventricular systolic pressure (LVSP), intraventricular pressure maximum upstroke velocity (+dp/dtmax),intraventricular pressure maximum decrease velocity(-dp/dtmax), heart rate (HR), coronary flow (CF),to explore the impact of CE on cardiac function. Results show that the CE on the isolated heart functions as two-ways regulation, when the concentrations of CE is lower than640μg/L, it can significantly improve the LVSP,+dp/dtmax,-dp/dtmax, showing the effect as a positive inotropic;when the concentrations of CE is higher than640μg/L,similar to the role of drug of verapamil, showing a significant negative inotropic effect,showing dose-dependent manner.(2) Using an isolated rat heart with ischemia-reperfusion injury to observe the impact of CE-related indicators of hemodynamic parameters, myocardium creatase, myocardial tissue structure to explore the protective effect on ischemia-reperfusion myocardial injury.We also use CE as samples to bulid the Langendorff model of isolated heart with myocardial ischemia-reperfusion injury,we observed LVSP,+dp/dtmax,-dp/dtmax,HR,CF; and we collected the coronary effluent after reperfusion30minutes,assaying the contenty of lactate dehydrogenase (LDH),creatine kinase (CK).At the end of the experiment, we cut the part of the left ventricle and put them into formalin, used HE staining and observed the the ischemia-reperfusion injury and degreed the pathological change of those injury.The results show that CE can reduce the content of the coronary effluent LDH,CK; and it can significantly reduce the degree of myocardial injury. Its hemodynamic parameters showing the two-ways regulation, When the concentrations of CE are lower than640μg/L the LVSP,+dp/dtmax and-dp/dtmax can be raised significantly,the drug can also slow the heart rate,increase the coronary flow in a dose dependent manner,the study also showed that the CE may increase the myocardial contractility while decreasing myocardial oxygen consumption,therefore it can reduce the extent of myocardial ischemia and reperfusion injury; When the concentrations of CE are higher than640μg/L,the heart rate were slower than the model group,+dp/dtmax,-dp/dtmax,LVSP showed a downward trend as well,its effect is similar to verapamil(20μg/L).The mechanism of CE is still need further studied.
