In order to realize the industrialization development and comprehensive utilization of acorn resource, the study was conducted on both removal process of tannin in acorn with ultrasonic and purification process of tannin in acorn with macroporous resins in this paper. In the removal process, two methods was compared in aspect of the removal effect of tannin under respective excellent condition of ultrasonic assisted water and ethanol firstly using the single factor experiments of the same factors settings. Meanwhile, the method of higher removal rate was chosen, and the orthogonal tests were designed to further optimize the removal process. In the purification process, firstly, through the investigation of various factors affecting the purified effect, the optimum purified conditions were determined. In addition, the validation test of best purification process was conducted.The obtained conclusions were as follows:(1) Ultrasonic assisted water and ethanol this two methods was used to remove tannin in the acorn, under their better removal conditions determined by single factor experiments of the same factors settings, the removal quantity of tannin were56.71mg/g and65.96mg/g, respectively, From removal effect perspective, ultrasonic assisted ethanol method was better than the ultrasonic assisted water method, obviously.(2) Under the better ultrasonic assisted ethanol method, the removal process of tannin in acorn was optimized by orthogonal experiments, the best removal process conditions were obtained after analysis of the experiment results:ethanol concentration was40%, particle degree was more than80yards, ratio of liquid to solid was1:30, ultrasonic time was60minutes, ultrasonic temperature was55℃, ultrasonic power was400W. Under the optimum conditions, the average removal quantity of tannin was69.99mg/g, removal rate was90.54%.(3) Through the investigation of various factors affecting the purified effect of NKA-Ⅱ type macroporous resins, the best purified conditions of tannin in acorn was determined: Sample concentration was4.39mg/ml, sample velocity was lml/min, sample volume was60ml, elution solvent was60%ethanol, elution velocity was lml/min, the volume of elution solvent was135ml.(4) Under the determined optimal process conditions, NKA-Ⅱ type macroporous resins were used to purify tannin in acorn, after purifying, the transfer rate of tannin was91.21%, the purity of tannins dry powder was74.08%.
