Microbes with all sorts of plants form symbiotic, then becomeendophytic microorganisms of the plant. The endophytic microorganismsspecies and biological characteristics are diversity, the endophyticmicroorganisms and plants form symbiotic relationship for a long time,the host to provide nutrition for the endophytic microorganisms, someendophytic microorganisms also improve the ability of host insects anddisease prevention.The endophytic microorganisms have many kinds ofbiological function, as some endophytic microorganisms may also helpthe host nitrogen fixation, promote the growth and so on. The endophyticmicroorganisms was found and focused as a kind of importantmicroorganism resources.The Camellia sinensis var. assamica was used as research materialin this study. Found the endophytic microorganisms of the Camelliasinensis var. assamica, these endophytic microorganisms from where,correlate with the microorganisms during fermentation of pu-erh tea. Theendophytic microorganisms and air microbes of Camellia sinensis var.assamica garden was identified simultaneously, then correlate with themicroorganisms during fermentation of pu-erh tea, determined involvedin the sources of microbes during fermentation of pu-erh tea primarily.The fresh and health leaves of Camellia sinensis var. assamica wassampled and dealed. Cultive the endophytic microrganisms from leaves.Using molecular biology methods Polymerase Chain Reaction (PCR,Polymerase Chain Reaction) amplification of16srdna sequences frombacteria or the ITS sequences from fungi, then amplified by cDNAsamples send to gene sequencing, according to the sequencing result,finds out the corresponding microorganisms from NCBI BLAST system,complete species identification. Different season, different degree of freshleaves and different varieties of he Camellia sinensis var. assamica leaves endophytic microorganisms was studied. Combination with the airmicrobiology research, to find the source of endophytic microorganismsof the leaves of Camellia sinensis var. assamica and combination withmicrobes during fermentation of pu-erh tea, to find endophyticmicroorganisms where to go.Sixteen kinds microorganisms which the bacteria are dominantwere isolated from Camellia sinensis var. assamica leaves in thispaper.The results show that endophytic microorganisms of Camelliasinensis var. assamica in buds and old leaf is less, more in young leaveswhich grow faster, endophytic microorganisms of Camellia sinensis var.assamica is not depend on variety. the endophytic microorganisms is themost in grow faster leaves which is pu-erh tea we usually drink rawmaterials.During the processing of fresh leaves to pu-erh tea rawmaterials, there are still some endophytic microorganisms along with thisprocess. We will also identify the microorganisms of pu-erh tea rawmaterials, to determine the endophytic microrganisms of fresh leavesexist in pu-erh tea raw materials and participate in pu-erh tea pilefermentation. The microbial species of Camellia sinensis var. assamicaleaves is regular. The microbial species of leaves associated withairwhich is the main source, some endophytic microorganisms involvedin pu-erh tea pile fermentation or along with pu-erh tea’s life.
