With growing social development and the increase in the world’s population, the human raceis faced with increasingly serious environmental crisis and energy crisis, cellulose is the mostabundant renewable resources, China’s annual crop straw, other than a small portion which arereturned to field, are all being wasted or incinerated, causing serious environmental pollution.Cellulose material handling methods are physical methods, chemical methods and biologicalmethods, but the absence of effective degradation, needs further study. So screening with theactive cellulase straw degrading microbial strains and related research are still currently hot anddifficult.This paper to develop efficient strains for research goals, as bacteria source of saline-alkalisoil in Daqing, sodium carboxymethyl cellulose as a carbon source, under the conditions of28℃,120r/min shaking culture, domestication, screening, isolated to obtain four advantages degradationbacteria, named A, B, C and D respectively. The four strains of fast-growing activity of secretioncomplete hydrolysis of lignocellulosic enzymes. By morphological observation, physiological andbiochemical experiments to combine16SrDNA homology and systems phylogenetic tree analysisidentified sequence analysis, preliminary identification of four bacteria the debauchery Penicillium(Penicillium Vinaceum), named A.sweet potato pathogenic fungus (Hypocrea sp.), named B, whiterot fungus (Hypocrea lixii) named C, linkage spore fungus (Alternaria sp.), named D.This experimental studied under different temperature, pH and substrate concentration valuesof four bacteria growth characteristics. Result that A bacteria at25℃experienced optimumgrowth, while B, C, D bacteria growth optimum temperatures were15℃, belonging to thecryogenic strains, making them Suitable for growth in the northeast region. Four bacterial genusacid bacteria, under the condition that pH is less than7, experienced optimal growth. Cellulosedegrading bacteria A under the condition that substrate concentration of3g/L, experienced themost vigorous growth, cellulose degrading bacteria B, C under the conditions of substrateconcentration of5g/L, experienced cell mass growth, reaching maximum. Cellulose degradingbacteria D in the substrate concentration of7g/L reaches a maximum in growth. They are arrived atthe highest value of0.0035g,0.006g,0.006g,0.007g. The characteristics of these four bacteriawere accurately understood. While Filter paper activity was carried out for these four bacteria,endo-glucanase activity and exo-glucanase activity determination. Cellulose degradation bacteriaA enzyme activity showed better results than the other three.This study shows, through studying the characteristics of cellulose-decomposing bacteria A, B, C, D, and by mastering its growth characteristics, we can improve cellulose degradationbacteria degradation efficiency. This is a way to the development a better utilization of crop stalksand other cellulosic resources.
