At present, the food and drug problem has become the focus of attention at home and abroad, as one of the important of the food and drug, plastic packaging materials safety is also introduced.reports from both at home and abroad, the damage of the plastic packaging materials mainly is the material inside of poisonous and harmful chemical pollutants migration and dissolution caused by contamination of food and drugs, which kind of chemical pollutants is plastic processing additives, processing AIDS are added the performance of the modified plastic, but it has different degree migrate to the food and drug, plus additives or its degradation may have certain toxicity to human body, so the processing aid could constitute a safety hazard of packaging materials, commonly used processing AIDS are:plasticizer, stabilizer, antistatic agent, antioxidant, etc. This article selects one of the agents-antioxidants, at present the commonly used four antioxidant (BHT, Irganox1010, Irganox1076, Irgafos168) as the research object, carries on the toxicology research,It provided the scientific basis for the future in-depth understanding of these for antioxidants for people,and it also provided nedessary technical guidance for reducing the antioxidant substances harmful to human.Random purchase commercially available seven kinds of food and drug packaging materials samples for this experiment, after processing and reflux extraction,the sample is analysed by high performance liquid chromatography(HPLC). result:the antioxidant content is different in different kinds of plastic packing materials or the same type of packaging materials, but it has a common is that BHT, Irganox1010, Irganox1076,Irgafosl68is contained in the most food and drug packaging materials, and Irganox1010, Irgafos168both content is higher, according to the results,It can be concluded that the contents of four kinds of antioxidants ratio is approximately (BHT:Irganox1010:Irganox1076:Irgafos168)1:6:1:9.With the compound (BHT:Irganox1010:Irganox1076:Irgafos168=1:6:1:9) as subjects to study the toxicology experiment, it respectively from acute toxicology test,genetic toxicology test,micronucleus test of born marrw in mice,sperms shape abnormality test in mice and30d feeding test to research the mixture toxicology. Results:the oral LD50of four antioxidant mixture (BHT:Irganox1010:Irganox1076: Irgafos168=1:6:1:9) is11340mg/kg BW; the results of micromucleus test of born marrow in mice and sperms shape abnormality test in mice were negative;the30d feeding test in rats demonstrated that it had no obvious toxic effects on growth and development,routine blood,biochemical index,VSI and histopathological changes. According to the classification of acute toxicity,the mixture of four antioxidants was classified in the non-toxic levels,and there was no damage to the chromosoma and sperm,as well as no significant subacute toxicity.Inrespectively Irganox1010,Irganox1076,Irgafos168as subjects,and from acute toxicity test and30d feeding test to sdudy its toxic effect. Result:the oral LD50of the three antioxidants were more than10000mg/kg. the30d feeding test in rats demonstrated that it had no obvious toxic effects on growth and development,routine blood,biochemical index,VSI.But in histopathological examination found that antioxidant1076group,168group,1010group rats renal tubular epithelial cell swelling, malpighian tube with tubular cells, and Irganox1010groups of rat intestinal mucosa inflammatory cell infiltration, can be concluded that when the concentration of800mg/kg dose, three kinds of antioxidants are subacute toxicity, and its target organs are kidney, besides, the antioxidant1010target organs also is intestinal.According to the acute toxicity classification standard,the mixture of four kinds of antioxidants, Irganox1010, Irganox1076, Irgafosl68can be judged to be the actual non-toxic according its LD50more than10000mg/kg. Under the condition of the present study, the mixture of four kinds of antioxidants has judge initially no damage to chromosome and spern accourding the result of mice bone marrow micronucleus test and sperm teratogenic test. Accourding the experiment result,the four kinds of antioxidant compounds without subacute toxic effect to rat,and the Irganox1010, Irganox1076, Irgafos168have subacute toxic effect on kidney of rats by preliminary judgement, and antioxidant1010has also subacute toxic effect on intestines.Its chronic toxic effect still need chronic toxicity test to further research.
