As a kind of testing technique, Raman spectroscopy can obtain the composition information and the molecular structure of the sample, with the characteristic of high sensitivity and non-invasive. At present, Raman spectroscopy has been widely applied in many fields, such as biology, chemistry and environment and so on. With the development of science and tecchnology, the Surface-Enhanced Raman spectroscopy (SERS) had been discovered and developed rapidly, it expands the application range of the Raman spectroscopy.This paper introduces the basic theroy and the application analysis technology of Raman spectroscopy, specifically discusses the enhancement mechanism of SERS. Active substrate is the key of SERS,It’s important to design new ones on the purpose of exploring the enhancement mechanism,so that SERS can be better used in the field of science and life.In the experiment of preparing silicon nanowires(SINWs) by the method of chemical wet etching, we found that the length of the nanowire would increase with the etching time goes,and the number would reduce after it increased to a certain value.As a kind of semiconductor nanomaterial, silicon nanowire can enhance Raman signal itself.This paple design a kind of SERS active substrate named Ag/SINWs,means silicon nanowires being coated with silver nanoparticles under the condition of Uv irradiation.Based on the prepared substrate, there are many "hot spots"on the external of the Ag/SINWs substrate so that the enhancement effect is good,the Raman spectroscopy of rhodamine6G (R6G) can be obtained in low concentration as10-9M. The Raman mapping study show that the Ag/SINWs substrate has good enhancement ability and good uniformity Detect the SERS of R6G with10-7M, during1to30days,the result indicate that the Ag/SINWs substrate stability under the condition of room temperature. So the Ag/SINW has good enhancement effect,and the SERS signal is almost uniformity and stability it’s a suitable type of SERS active substrate.
