The corn yellow serofluid is byproducts of corn starch in the industrial production.,the content of organicis high,governance will undoubtedly become more difficult,so the best way is to change waste intotreasure,and this is precisely in line with our environmental policy.The corn yellow serofluid contains lots ofnutrients,such as carbon and nitrogen sources.It can take advantage of some glucose as the carbon sourceinstead of medium in the production of food fermentation industry.It is significance to reduce the cost ofindustrial production and reduce the pollution caused by the byproducts of emissions on the environment andits use of resources wasted.Because of its unique physiological activity,arachidonic acid apply to manyareas.Because polyunsaturated fatty acid content is low and higher extraction costs in the animal and vegetableoils and fats,so it has been unable to meet the demand of the market.Microbial fermentation production ofarachidonic acid is particularly prominent at this time.This topic select Mortierella isabellina high-yield strainYZ-124,investigate the fermentation medium optimization adding to some content of corn yellowserofluid,optimization of culture conditions,oil extraction and purification, finally,using meteorologicalchromatography analysis the content of arachidonic acid. The specific results of the study are as follows:1.On the basis of the medium of the microbial fermentation production of arachidonic acid in thelaboratory,adding a certain amount of the corn yellow serofluid instead of a part of glucose in order to reducethe costs of industrial production.Determine the optimum fermentation medium: Initial glucose concentrationis90g/L,the amount of the corn yellow serofluid is25%, initial pH is6.In this optimum conditions themaximum yield of arachidonic acid is3.11g/L.2.On the basis of single factor tests,Orthogonal combination of testing to determine optimum cultureconditions:The temperature of the fermenter is28℃,fermentation time is7d,using50%of the fed-batchmode.In this optimum conditions the maximum yield of arachidonic acid is3.22g/L.3.Using the orthogonal experiment to optimize the process parameters of ultrasonic assisted extraction ofoil,and the optimum extraction process is:high-frequency,time is10min,temperature40℃,extraction times2times,ultrasonic time30min,materiar/liquid ratio is1:9.In this optimum conditions the maximum yield ofarachidonic acid is3.45g/L.Through weather chromatographic determine the content of arachidonic acid is24.15%.4.Mixing purified microbial oil using of urea inclusion centrifugal technology,ang using Plackett-Burmanexperimental,steepest ascent and central composite rotatable design and response surface analysis methods andmodel optimization,the best purification process parameters are:urea/fatty acid is1.7:1,the concentration ofethanol is98%,Centrifuge speed is3300r/min,under this condition,the actual content of the ARA can reach45.8%,improved than single purification of23.2%and than before purification of87.8%.
