iPS cells, known as induced pluripotent stem cells, can be derived from somatic cells when they are driven reversely back to embryonic stem cell-like cells by introducing several genes (Oct4, Sox2, Klf4, c-Myc, Nanog, Lin28). Except for the origin, iPS cells have the same characteristics as embryonic stem cells (ES cells):They have pluripotency which means these cells can differentiate into any cell type in theory.Two different types of pig cell have been used as experimental material for induced pluripotent stem cells in this study. First, by isolating, purifying and identifying from pig ovarian tissue, pig ovarian epithelial cell line has been established. Second, virus packaging of pMXs-mOct4, pMXs-mSox2, pMXs-mc-Myc and pMXs-mKlf4have been performed with Lipofectamine2000. Finally, by introducing four defined transcription factors into pig fetal fibroblasts and ovarian epithelial cells via retroviral transduction, the two types of cell have been induced into pluripotent stem cells and the reprogramming efficiencies were compared. The results show:pig fetal fibroblasts in younger passage have higher reprogramming capacity than those in elder passage; the reprogramming capacity of pig ovarian epithelial cells becomes better when these cells completely change to fibroblasts. This study provides some evidence for the choice of somatic cell. |
