| It is not only very important to spun the silk and knot the cocoon for existenceand defending natural enemies of silkworm.,but also the silk can be used by people andhas a great important action in the silk industry, trade, agriculture, and culturalinteractionCocoon is composed of frison, cocoon shell and chrysalis and shed skin.thecocoon shell can be used for capacity filature and cocoon shape is a main sign forclassifying the silkworm species,because of difference species, the cocoon shell isdifference.In the secular evolution process,silkworm backlog a great deal of mutation body andbreeding system, and mutant related cocoon characteristic mainly have flossy cocoon,sericincocoon,outside layer yellow cocoon, green cocoon, and coarse cocoon and so on,whice arethe important materials for studying the cocoon characteristic.The thesis will mainlyintroduce silk formation and cocoon behavior and mechanism of coloring cocoon relatedfoundation information,concretely as follows:(1) We apply two-dimensional gel electrophoresis to analyse different periods of the5th instar stage proteins from different section of the middle silkgland of3varieties withdifferent cocoon behavior which are normal cocoon,flossy cocoon and sericincocoon,respectively,and found there are some distinct protein spots among the proteinsextracted from the same period and section middle silk gland cells of three silkworm breeds.Matrix-assisted laser desorption ionization and time of flight mass spectrometry(MALDI-TOF-MS) was applied to identify some notable distinct protein spots,and itacquires thirty-five reliable protein identification results,which are blasted in the proteindatabase,involve in many biological processes such as energy metabolism and proteinsynthesis. serpinl6and serpinl8identified in this article may provide useful information forunderstanding of the formation mechanism of the flossy mutant of silkworm (Bombyxmori).(2) Reciprocal backcrossed F1(BC1)progenies are matched with silkworm strainsBaghdad(Ba),which express white hemolymph (+Y+Y II),and KY which express yellowhemolymph (+Y+YII),and they are called BC1F and BC1M respectively.Base on thepreliminary mapping of I gene in this laboratory, we search for the new molecule markersfor fine location of I gene.5SSR and1STS markers were found to be linked with Igene,Meanwhile, We constructed a linkage map of12.8cM and I gene WAS located between I001and STS48-1.The nearest markers is STS48-1,And the distance between Igene and the nearest markers STS48-1is0.5cM.
