Microsporidia are obligate intracellular protozoan parasites that infect a broad range of vertebrates and invertebrates,they were recognized as a cause of disease in many nonhuman hosts including bird, fish, insects, amphibian, mammals(dog, cat, pig, rabbit, sheep. et al.), and human. The focus of this review is to discuss the most recent perceptions on the zoonotic potential of the various microsporidia with proven vertebrate hosts.The first human case of microsporidial infection was reported in 1959, and they emerged as important opportunistic pathogens when AIDS became pandemic, only 35 cases of microsporidiosis have been documented. Microsporidia can infect immunocompromised patients and immunocompetent. but also in immunocompetent persons with acute, self-limited diarrhea.however, Immunocompromised especially AIDS patients may be resoult to lethality diarrhea, Microsporidia as new pathogens section was found, and is considered the cause traveler diarrhea worldwide. As people living standard rise ceaselessly, Microsporidia gradually attracted people’s att-ention.We usded to Chromotrope 2R stain, Qick-Hot Gram-Chromotrope stain and Trichrome-Blue stain to investigate Microsporidia from children in zhengzhou. The result showed that the Qick-Hot Gram-Chromotrope stain methods is better than others in the whole experiment. The stain method can distinguish the microsporidia spores, provide the basis of the epidemiology of microsporidia. In order to learn about the infection of intestines parasitic children in zhengzhou, We used to Saturated sugar floating, Iodine staining detect 1996 specemens of children from hospital in zhengzhou between July 2009 and July 2010 in Laboratory, the total infection rate 1.5%, the infection rate of Giardia, cyclospora cayetanensis was 0.6%, 0.5%. the infect of rate Cryptosporidium, amebic protozoa, Strongyloides stercoralis is 0.1%, 0.3% and 0.1%, The survey did not find mixed infection.In order to understand the popular information and morphological characteristic of Microsporidia in Henan Provice, We used to Saturated sugar floating, aldehyde-ether precipitation method and Quick-Hot Gram-Chromotrope 2R staing, the result shows tha 11 fecal specimens were diagnosed as Microsporidia positive.In order to determine species or genotype or subtype of Microsporidia isolates from children feces in zhengzhou. We used nested PCR assay to 392-bp region of ITS gene, 2 of 11 microscopy-positive samples from human and 4 samples from pig were amplified.Then, followed by DNA sequence analysis, Then, Blast methods search homological sequences in NCBI, after that, homological sequences were alignmented. Phylogenetic tree and homological analysis were made by some biological softwares such as Clustal X 1.81 and DNAstar4.0.
