Foxtail millet(Setaria italica L.)was one of the most ancient cereal crops in the world.As a staple food foxtail millet has made great contribution to sociaty development especially in China.Foxtail millet was characterized with its drought lerance,salt tolerance,infertile soil tolerance,highly efficiency of water utilization,relatively balanced necessary nutrition for human consummation and other special traits.Millet is the diploid flower crop pollination,its genome small(470Mb) and very stable very suitable to serve as research object genome.But millet is regional important crops,only grow in many developing countries and less developed countries,less research investment,cause millet genome and biotechnology research relatively lags behind other major crops.As the world population growth,the demand of food is also increases accordingly,this needs genetic engineering technology to improve the quality and yield of millet.The establishment of high efficiency,excellent millet transformation and regeneration system is the premise of genetic engineering methods selective breeding.This article analysis and optimization the influencing factors of callus induction using foxtail millet shoot apex as the explant,the callus of inflorescence is texture closely and has a lot white particles,but the green bud of callus formationed could not further growth and differentiation and not get the regeneration plant.The experiment of callus induction using foxtail millet shoot apex as the explant,transfer three varieties of millet into the medium with the concentration of 2,4-D was 2.25μmol/L、4.5μmol/L、9.0μmol/L.The result show that 2,4-D has a significant effect on callus induction.This article analysis the influencing of Kn and Zn,and Kn was suitable for callus induction.Sugar as the basic caibon of medium has some influence for callus induction,and sucrose was suitable.In addition we transformed the primary callus and embryos callus using Agro-bacterium tumefaciens-mediated,and conducted GUS staining analysis,the result indicated that the transformation of primary callus GUS transient expression efficiency was higher than embryos callus.This work will provides reference data for millet tissue culture and genetic transformation research in future.
