Syrmaticus humiae is classified as the animal of Phasianidae,which was listed as Ⅰ-class national protected animals.This species was distributed narrow and declined dramatically in china. Therefore, Syrmaticus humiae has been included in the "Appendix I" of the CITES. It has been designated as an Near Threatened species in the Red Data Book of the IUCN of China in2005.To save this endangered species,RAPD marker was carried out for discussing the genetic diversity of Syrmaticus humiae in this study. We have analyzed the genetic diversity of cultured stocks of Syrmaticus humiae and explored their phylogenetic relationships. The extent of the genetic variation reflect the potential evolution.Therefore,genetic diversity and its evolution is an important genetic indicators for conservation biologist,which is evaluated to the potential evolution of a species,as well as the ability to withstanding the various survival pressure from the nature. Understanding their genetic diversity,the purpose is to provide a scientific basis for cultured stocks of Syrmaticus humiae on protection and management. Eventually,the endangered pheasants can return to the nature and restore their wild populations.We hope that the near threatened pheasants can avoid the extinction threat and in the natural environment to survive and propagate.The genetic diversity of cultured stocks of Syrmaticus humiae was studied as revealed by RAPD marker.14from50random primers were screened for PCR amplification for stock genetics analysis.The amplified bands were clarity,and highly specificity.Before the official polymerase chain reaction (PCR),we have Choiced the reaction composition (template DNA,primers,dNTP,Taq enzyme etc) and reaction conditions,in order to enhance the stability of the RAPD-PCR reaction.The results of this study show that the stability of RAPD is better. Analysis of Syrmaticus humiae groups have found that:The average percentage of polymorphic bands (P) is0.8235,The Shannon diversity information index (Ⅰ) is0.4290. The Nei’s genetic diversity index (He) is0.2885.The average genetic distance of Syrmaticus humiae was0.2810. In other words,similarity coefficient of individuals within a population was0.7190. This show that stocks of Syrmaticus humiae is lower genetic diversity. UPGMA was used for cluster analysis for twenty-four individuals of Syrmaticus humiae,through calculating the genetic similarity coefficient between individuals and the genetic distance.24individuals were obviously divided into three groups.This result was illustrated that the population of cultured stocks of Syrmaticus humiae individuals from different populations.
