In this study, shoot induction, regeneration rate, sensitivity to kanamycin and transgenic frequencies of different explants transformation by Agrobacterium-mediated in soybean were analyzed. Transcription factor GmPHR1 and OsPHR2, which were involved in signaling of phosphorus starvation, were transformed into soybean genotypes via Agrobacterium-mediated and pollen tube pathway transformation method to obtain soybean germplasms. The main results were as follows:1. The shoot tip and embryonic tip systems were more ideal soybean genetic transformation systems than others in this study. The shoot tip transformation system were the highest shoot induction, shoot regeneration, transformation ratios and little genotype dependence. But it also were the least sensitive to antibiotics, so there were some escapes in this system. The embryonic tip transformation system had higher shoot induction, shoot regeneration, transformation ratios and sensitive to antibiotics.2. The transgenic soybean plants with GmPHR1 and OsPHR2 were obtained via Agrobactetium-mediated method. Over-expression vector GmPHR1 and OsPHR2 were introduced into soybean variety Ji-dou 12. PCR and RT-PCR analysis results showed that nine T4 plants(1 line)with PBI121-GmPHR1, 23 T2 plants(9 lines) with Px6-GmPHR1 and 78 T3 transformation plants (12 lines)with OsPHR2 were obtained.3. The transgenic soybean plants with GmPHR1 were obtained via pollen tube pathway transformation method. And the soybean varieties Zhong-huang 41, Sui-nong 14, Ji-dou 3 had higher podding rates than other varieties . Two T2 positive plants (1 line) of Ji-dou 12 and four T2 positive plants (4 lines)of lv 75 with Px6-GmPHR1 were also obtained. And seven hundred and six transformation seeds with GmPHR1 and three hundred and fourty-five transformation seeds with OsPHR2 were obtained.4. Under the phosphorus starvation condition, the tolerance of transgenic soybean plants with OsPHR2 was better than wild-type control. In addition, the hundred-seed weight and seed protein content in transgenic soybean increased by 3.25% and.1.05% respectively.
