Genetic Polymorphism Of GDF9, BMP15, BMPR-IB, OPN Genes And Their Correlation With Reproductive Traits In Pig

In this study, the polymorphism of4candidate genes including GDF9, BMP15, BMPR-IBand OPN gene in Landrace pig, Large White pig, Duroc and Huai pig were analyzed byPCR-SSCP or PCR-RFLP, and association analysis of their polymorphisms with reproductivetraits (Total number born, TNB; Number born alive, NBA) were analyzed by parity, involvingthe first parity and the later parity (from the second parity to the eighth parity). Theresults as follows:10mutations were detected in4genes, they are including G530A and C2733T in GDF9gene,T503G and A5471T in BMP15gene, A1081C, A14990G and T19588A in BMPR-IB gene,T325G, C6140A and C7133T in OPN gene.3genotypes were detected in these10mutations,respectively.Association analysis of litter size with polymorphism of G530A mutation in GDF9geneindicated that TNB and NBA of both Landrace pigs and Large White pigs with AA genotype weremore than those of pigs with GG and AG genotype (AA>GG>AG), while TNB and NBA ofLandrace pigs with AA and GG genotype were more than those of pigs with AG genotype for thelater parity (P<0.05); In both Duroc pig and Huai pig, AA genotype was not found, while pigs inthese2populations (both first parity and later parity) with GG genotype showed more TNB andNBA than those with AG, but there were not significant differences (P>0.05).Association analysis of litter size with polymorphism of T503G mutation in BMP15geneindicated that TNB and NBA of Landrace pigs, Large White pigs and Duroc pigs with TTgenotype were more than those of pigs with TG and GG genotype (TT>TG>GG), while TNB andNBA of pigs with TT genotype were more than those of pigs with GG genotype significantly inthese3populations for the later parity, respectively (P<0.05). In Huai pig, TNB and NBA of pigswith GG genotype were more than those of pigs with TT and TG genotype (GG>TT>TG), whilepigs with GG genotype showed more TNB than pigs with TT and TG genotype significantly forthe later parity (P<0.05). Association analysis of litter size with polymorphism of A1081C mutation in BMPR-IB geneindicated there were not significant differences in TNB and NBA of pigs in these4populations forthe first parity. In the later parity, there were trends of AA>CC>AC in TNB and NBA ofLandrace pigs, Large White pigs and Duroc pigs, TNB of Large White pigs with AA genotypewere more than those of pigs with CC genotype (P<0.05), NBA of White pigs with AA genotypewere more than those of pigs with AC genotype (P<0.01), NBA of Duroc pigs with AA genotypewere more than those of pigs with CC genotype (P<0.05). Huai pigs with AC genotype showedmore TNB and NBA than those with AA and CC genotype for the later parity, but there were notsignificant differences (P>0.05).Association analysis of litter size with polymorphism of T19588A mutation in BMPR-IBgene indicated that TNB and NBA of Landrace pigs with TA genotype were more than those withTT (P<0.05) and AA genotype(P>0.05). There were not significant differences between differentgenotypes for both Large White pig and Huai pig in TNB and NBA (P>0.05), but they with trendsof TA>AA>TT. Meanwhile this mutation was not found in Duroc pig, and only TT genotype wasdetected.Association analysis of litter size with polymorphism of T325G mutation in OPN geneindicated that pigs with TT genotype and TG genotype showed more TNB and NBA than pigswith GG genotype significantly in Huai pig for the later parity (P<0.05). Besides, there were notsignificant differences between different genotypes for Huai pig (first parity) and other3populations (both first parity and later parities) in TNB and NBA.Association analysis of litter size with polymorphism of C6140A mutation in OPN geneindicated that no significant differences were found between different genotypes for Landrace pigand Large White pig (both first parity and later parities) in TNB and NBA (P>0.05). There areonly AA genotype was detected in Duroc pig. In Huai pig, pigs with CC and AA genotype showedmore TNB than pigs with CA genotype for the first parity (P<0.05), while there were notsignificant differences between different genotypes in NBA for the first parity (P>0.05), TNB ofpigs with CC and AA genotype were extremely more than those with CA genotype for the later parity (P<0.01). NBA of pigs with CC genotype were extremely more than those with CAgenotype for the later parity (P<0.01), while NBA of pigs with AA genotype were more thanthose with CA genotype for the later parity (P<0.05).Conclusion was drawn that the G530A mutation in GDF9gene, the T19588A mutation inBMP15gene and the T325G mutation in OPN gene can’t be determined whether they areassociated with reproduction traits in pigs. While the T503G mutation in BMP15gene, theA1081C mutation in BMPR-IB gene and the C6140A mutation in OPN gene maybe associatedwith reproduction traits in pigs, and increasing the litter size of pig is possible by using MAS.
