CaM is thought to play an important role in the calcium metabolism involved in shell and pearl formation. But by now, the specific fuction of CaM in the process of calcium metabolism in Hyriopsis schlegelii is still uncertain. In this rearch, we searched for the expression and localization for CaM, and hope that we can provide some advice and guidance for pearl production.The result of QRT-PCR showed that the CaM mRNA expression level have3peaks during the research time.The8th day after nucleated operation, the CaM mRNA expression level showed sharp up-regulation in pearl sac, however, the8th day after non-nucleated operation, the CaM mRNA expression level showed sharp up-regulation in mantle tissue. Besides, at the120th day after the two operations, the CaM mRNA expression level of the pearl sac tissues both have up-regulation. The CaM mRNA expression level after nucleated and non-nucleated operation were both under that of the mantle of contrast group.The complete ORF of HsCaM subcloned into PET-28a prokaryotic expression vector, with affinity chromatography purified expression products, use the products as antigen to immunize rabbit, polyclonal antibody was successfully prepared for immunofluorescence experiments. The results of immunofluorescence experiments showed that HsCaM mainly expressed in the epithelial cells of mantle, proliferating outer mantle epithelial cells of the mantle graft and the epithelial cells of complete pearl sac.
