Selenium (Se) is an essential dietary component for animals and humans. Though there isno report about Se is an essential micronutrient for plants, it has important biochemical effectsfor plants’ development, especially its role of antioxidants and antioxidant-related enzymes inprotective response to adverse environment. This research was to study the possibility ofgrape cultivated for enriching Se and the protective effects of Se with grape cutting seedlingsunder water stress. The aim of this work was to offer some theoretical guidance for the futureresearch about enriching Se with grape and drought resistance cultivation of grape. This studyhad two parts, field experiment and pot experiment. The V. vinifera L. cv. Cabernet Sauvignoncultivated for6years was used as material. In field experiment the materials were fertilized insoil with different sodium selenite conten (22.5mg/plant、45mg/plant、67.5mg/plant) ingrape flowering stage and also sprayed with different concentration of sodium selenitesolution (50mg/L、100mg/L、150mg/L) to all plant in the first berry expansion stage. Inorder to study the effects of Se-ferlilizer under two different treatments on grape quality. TheV. vinifera L. cv. Cabernet Sauvignon cutting seedlings cultivated for two years were sprayedwith different concentration of sodium selenite solution(0.5mg·/L,1.0mg·/L,1.5mg·/L)under water stress, which was used to analysis the effect of Se-fertilizer on grape droughttolerance. The main results of the experiment are as follows:1. Under two different Se-fertilizer treatments, the more dose with Se-fertilizer treatedthe more Se enriched in grape berry and leaf. Moreover, Se enriched in grape leaf was muchmore than berry, and the absorption effect of spraying treatment was better than soilfertilization.2. The treatments of lower level of Se content (22.5mg/plant、45.0mg/plant) in soil andall spraying fertilized treatments can enhance the reducing sugar content of CabernetSauvignon during maturation. However, soil application of larger amount of selenium (67.5mg/plant) may inhibit reducing sugar increased in grape maturation. On the countrary, Sefoliage spraying treatment can decrease total acid.So it can improve grape maturation in allmaturing stage.3. Se-fertilizer treated with spraying or in soil can also promote total phenolic and total anthocyanin content in grape skin, and also can improve the scavenging ability of ABTS+,DPPH and hydroxyl radical in all maturing stage. These two treatments can also improve themetabolization of tannin during grape maturation and can promote the grape maturity index.With comprehensive analysis, the spraying treatment with100mg/L sodium selenite solutionis much better to improve the grape quality.4. With pot experiment under water stress, exogenous selenium with appropriateconcentration can encourage the content of soluble sugar, soluble protein and proline of grapeleaf, and can also improve the activity of POD, CAT and GSH-Px, and reduce the MDAcontent of membrane lipid peroxidation as well. While it had negtive effect on SOD activity.5. Under water stress, the application of exogenous selenium with suitableconcentration can mitigate the RWC decline and chlorophyll degradation of grape leaf, andthis effect was more clearer with Se concentration increasing. While under the same waterstress degree the treatment of lower Se concentration(0.5mg/L) can increase the Pn, Tr, Gsand Ci of grape leaf, but it can decrease photosynthetic indicators with higher concentrationSe treatment (2.0mg/L).6. Under this experimental condition, considering all these biochemical indexes ofCabernet Sauvignon seedling leaves under water stress,, the best effect of sodium selenitesolution to alleviate the damage of drought on grape was spraying treatment with1.0mg/Lsodium selenite solution.
