Food is the foundation of human survival and development. Rice is one of the world’s most important grain crops and nearly half the world’s population live on the rice for a living. So it is extremely important to ensure good quality and high yield of rice. Sufficient moisture is the basic condition for normal growth and high yield of rice. However, drought is a common problem worldwide. Drought seriously constrains the normal growth and development and yield formation of rice.Drought resistance is controlled by multiple genes, that is why using conventional breeding to improve crop drought resistance is limited. At present, the popular research mainly concentrates on using resistance genes to improve crop drought resistance at home and abroad. Recent progress has been made in under standing of gene transcriptional regulation and signal transduction in plant responses to drought on the molecular level. It is in this context that drought-resistant research of rice has become a hotspot and focus of the current scientific research.AK07gene, a drought tolerance gene of rice, is screened by using gene chip technology. Constructing over expression vector of the purpose gene and making it expressed in the transgenic plants. It can be preliminarily confirmed that AK07gene has drought resistant function by observing the different phenotype of transgenic plants. To better understand the role of AK07during drought tolerance mechanism, AK07gene has been isolated from rice and its expression patterns and functions were analyzed. The main results and conclusions are as follows:(1) Constructing the over expression vector and made it transformed into rice: PCR confirmed that foreign gene had already integrated into the genome of rice. Drought simulation test and drought test showed that AK07gene could enhance drought resistance of the rice.(2) Constructing the analysis vector of the promoter and made it transformed into rice:The analysis of Gus activity indicated that the expression was detected in the leaves and internodes during seedling stages and during booting stages it was detected in the spikelet, especially in the anther and lemma. The result of paraffin section experiment further suggested the definite part which gene AK07had mainly expressed in.(3) Constructing fusion expression vector:The result of sub-cellular localization showed that the AK07gene was expressed on the outer wall of cells.(4) Constructing interference expression vector and made it transformed into rice: The transgenic plants had been acquired.
