Euphorbia kansui Liou which belongs to Euphorbia in Euphorbiaceae is a perennial herbaceous species. They are mainly distributed in shaanxi, henan and gansu provinces. Their dry roots have been used as a traditional Chinese medicine. The medicinal value is very important and they have antiviral and anti-cancer properties. Diterpene esters are the effective components of Euphorbia kansui and these medical compositions are accumulated in milky sap. At present, most studies about E. kansui had mainly focused on their chemical components analysis, pharmacology and clinical applications. As far, proteomics research of Euphorbia kansui has not been reported. As a result, at present study, SDS-PAGE and two-dimensional gel electrophoresis (2-DE) methods were firstly used to separate proteins in Euphorbia kansui milky sap and then to identify some milky sap proteins using HLPC-MS and MALDI-TOF-MS.1. SDS-PAGE protein profile of milky sap extracted from Euphorbia kansui showed11protein bands. The molecular weight of most of the proteins was30-66kDa, and there had high-abundance proteins in66kDa.2.58proteins were identified by using HLPC-MS, which comprised proteins involved in the development of laticifers, disease/defence-related protein and proteins involved in general metabolism.3. The protein difference of Euphorbia kansui milky sap collected respectively from laticifers at the early and mature developmental stages was compared by two-dimensional gel electrophores.19proteins whose content had two or more times differences were indentified using peptide fingerprint mass spectrometry technique. Their possible functions in the development of laticifers were also analyzed.
