260and168fecal samples of wapiti(Cervus elaphus xanthopygits) were respective collected in Wupao Farm the Wandashan Mountains from February to March in2010and February to April in2011. The DNA extraction from fecal by the QIAmp DNA Stool Mini Kit, and then the Polymerase Chain Reaction、STR and Sex Test using9polymorphism loci. This study extracted HIQ DNA from fecal using the microsatellite DNA to analysis the deer genetic structure、inbreeding、inbreeding depression、 inbreeding avoidance、subpopulation dynamics and the home range.1、The results show that33different wapiti individuals from260fecal samples, and21females,12males in2010, and then19different ones from168samples,10females and9males in2011. Meanwhile,16identical individuals were observed.2、10parentage groups were obtained by the parentage analysis of the Cervus3.0.and two of them were inbreeding in2010, and5parentages and1inbreeding in2011.3、7subpopulations and8separate individuals were collected in2010, and4subpopulations and8separate ones in2011by Arcgis9.3and record in field work. But more, females and males mixed subpopulations were common and females groups or males ones were rare in2010and2011, the migration of subgroups and separate ones was frequent, especially females.4、We calculated the home range of wapiti in2010and2011. The average area of females was169.28hm2, and males was254.18hm2, the male’s area was1.6times female’s. The change that home range of the same individual was increased680.6%maximally due to heavy snow in2011. The average area of subgroups was686hm2and621hm2, reduced9.7%by contrast.5、70%kernel is fit for the home range of subgroups and50%kernel for centre range, meanwhile52.47%area in the study fields is appropriate for wapiti’s living and24.8%is best fit. The overlapping home range of fathermotherchildren is common, but the migration distance is inconspicuous.6、The dynamic integration of Fecal molecular biology and3S technology is obvious advantage to the Radio-tracking Technology.
