Atractylodes lancea (Thunb.) is a kind of herbaceous plant in Asteraceae family. As the original plant of Chinese herbal medicine Atractylodes lancea,according to Chinese Pharmacopoeia(2005).The dried rhizome is used as medicine, which is traditional medicine blindly to medicine。It’s warm and benefit for spleen,stomach, liver,which has an effect of strengthening spleen,clearing away heat,,removing wind and dispersing cold.In recent years,the market supply mainly rely on artificial cultivation, because of the wild resource is shortage. In the field production, rhizome is used as the propagation material usually.At the same time,seed renovation and new varieties breeding will be demanded. The reason for this is the vacancy of Seed testing rules and The seed(seeding) quality standard.Consideration of lack of Atractylodes lancea growth research, the study on Growth Regularity of A.lancea, Including the phenophase division, dry matter accumulation and distribution,medicinal extract content comparison in different picking period of A.lancea.,leaf length to width ratio, photosynthetic comparison of A.lancea from different populations. In this study, we discuss indicators of the testing method,based on the characterization of Atractylodes lancea seed quality.Combined with field cultivation test results,we discuss some related index of seed and seeding,then formulate the seed and grading standard of A.lancea finally.It lay a foundation of resource evaluation and good variety selection of A.lancea,and provide reference for guidance of Atractylodes lancea standardization production.The specific results:1. In the process of growth and development of A.lancea,the growth of leaf dry weight is rapid,other parts is lower in April,.From May to July, vigorous growth, the part on the ground mainly.In July,flower bud and plant turn into the reproductive growth stage.The part on the ground (leaf especially) slowdown, underground part begins to increase significantly.In September, the ground part cease growth or fall over basically,underground portion has maintained a high growth rate. The result equates with the other literature reports. 2. The content of solute in water and ether soluble,increased progressively from September to October, until late October reaches a maximum value, then reduce gradually. The solute in alcohol only is highest in September.3. Difference of Atractylodes lancea leaf length to width ratio From Different populations is apparent, followed by Suizhou, Nanzhang, Yunxi, Yichang, Yingshan, Shaanxi, Jurong, Shennongjia, Danjiangkou and Dawu. The internal variability of leaf length to width ratio from Different populations in size is in sequence of Suizhou, Nanzhang, Shennongjia, Jurong, Yichang, Yunxi, Danjiangkou, Yingshan, Shaanxi and Dawu.It illustrate the plant traits vary greatly in different populations and the same area.4. The origin of plant photosynthesis efficiency from high to low is in sequence of Dawu,Yingshan, Nanzhang, Yunxi, Shennongjia, Yichang, Suizhou, Shaanxi, Jurong and Danjiangkou. The origin of plant blame WUE from high to low is in sequence of Nanzhang,Yingshan,Dawu, Shennongjia, Suizhou, Yunxi, Yichang, Shaanxi, Danjiangkou and Jurong. There were differences in capacity of photosynthesis and water use ability in different populations.5. Determine the unarmed halving method is the optimal method of seed sampling.The unarmed method and7mesh sieve screening are the feasible methods of seed purity determination.The method of five Hundreds particle is the optimal method of seed1000-seed weigh determination.The seed moisture determination conditions are grinded seed as pretreatment,drying120min in120℃.The seed viability determination conditions are staining for3h in40℃,1%TTC solution.With the perlite as germination bed,22±1℃and60%-80%saturated moisture condition on Seed Germination.By direct inspect method, break method, staining method and proportion method of seed insect infectiosity determination. Condition of agar as a substrate and maintain30℃culture is selected to seed disease infectiosity determination. 6. The germination rate of The-one-standing seed is not less than89%,1000-grain weight is not less than10.4g, the viability is not less than90%.The germination rate of the-two-standing seed is84%-89%,1000-grain weight is8.6g-10.4g, the viability is84%~90%.The germination rate of the-three-standing seed is79%-84%,1000-grain weight is6.4g-8.6g, the viability is74%-84%. The seed purity is not less than85%, water content is less than9%,pest and disease infectiosity were less than7%in all grades of seed.7. The rhizome weight of the-one-standing seeding is not less than10g, the-two-standing seeding is5.6g-10g, the-two-standing seeding is from1g to5.6g.
