Corn is one of the world’s three major food crops, genetic transformation is one of the most efficient methods to increase maize yield and quality. Successful genetic transformation depends primarily on the excellent acceptor system. Compared with immature embryos, the mean explants for transgenic acceptor system of maize, multiple buds form mature embryos are available year round and in bulk quantities. It also have high frequency induction and the regeneration plants have no variations. How to increase the frequency of multiple buds induction and regeneration is the aim of this research. The mainly results form the experiment as follows:1. Using CML295as the acceptor material, randomized block design of experiment which contains8different induction media with different concentration of6-BA and2,4-D. Each medium includes3repeat trials, and use the induce frequency to make the analysis of range. The highest frequency of induction is about63%.2. Research the subculture and Propagation of the multiple buds induced by3.5:0.5and3.0:0.5induction media. The multiple buds induced by3.5:0.5induction medium can subculture6months compares the3.0:0.5induction medium with16months.3. Analyse the quality of multiple buds induced by different induction media and create an evaluation criterion of quality of multiple buds. Search out the most appropriate induction media to induce multiple buds is3.2:0.5inductive medium.4. Observe the samples of the multiple buds in different growth stages by scanning electron microscope.5. After the induction, differentiate and regenerate the multiple buds which have been subcultured over2,4and6months. As the result, the differentiate ratio of2moths subculture multiple buds is over80%,50%for4months subculture multiple buds and0for6months. At last, the seeding rate of2months subculture multiple buds reaches60%.
