The yellow peach moth, Conogethes punctiferalis (Guenee)(Lepidoptera:Crambidae), is one of the most serious pests that distribute widely in the world. There might be genetic differentiation due to different hosts and large extent distributions. In this research we collected yellow peach moth from different geographical populations of China, including Liaoning, Shandong, Guangdong Hebei, Henan, Hubei, Sichuan, Zhejiang and Beijing, living on corn, sorghum, sunflower, chestnut and masson pine, respectively. To analysis if there are high genetic differentiation between populations, we analyzed the gene flow and genetic differentiation between different geographical populations, and estimated genetic dynamic. In this study, we also investigated the prevalence and diversity of Wolbachia in the yellow peach moth. The results were as follows:1. From the phylogenetic analysis we found that there were significant differences between the pinaceae-feeding type (PFT) and fruit-feeding type (FFT), our study set consists of morphological data and two mitochondrial genes giving strong support for PFT and FFT having segregated into different species statuses of Conogethes.2. The sequences showed a high polymorphism in mtDNA of the yellow peach moth. The number of haplotypes of the three genes (COI、COII, Cytb) were116、53、122respectively. The evolution rate of these three genes were evaluated using polymorphism: Cytb>COI>COII.3. The24geographical populations were divided into three groups in order to study the genetic differentiation and gene flow within and among these groups. The group of south area (Guangdong and Guangxi province) showed a minimum gene flow within the populations of this group and among other groups. Similarly, the highest degree of genetic differentiation were visualized in the phylogenetic tree. However, the anlysis of molecular variance (AMOVA) results revealed that most of the genetic variation was within populations.4. The mismatch distribution of three genes in the yellow peach moth populations showed that there might been a population expansion in history, during14990-274450years to date.5. The average rate of Wolbachia infection was2.56%, which showed extremely low infection rate in yellow peach moth in China. The Wolbachia has many types and distributed mostly in south and southwest of China.8strains were detected in this study, but the cluster tree showed no remarkable distributed structure with their hosts of different geographical populations. It is the first time to combine morphological data and three mitochondrial genes to settle a long-standing question on the position and rank of yellow peach moth. It is proved to be a good evidence for the deduction that fruit-feeding type (FFT) and pinaceae-feeding type (PFT) were different species. The haplotypes of three genes (COI、COII、Cytb) of Conogethes punctiferalis were submitted in the GenBank to share the sequence data with entomology of worldwide field. Also, it is the first time to analyze the genetic differentiation and population dynamics, to discuss the gene flow and the differentiation, to draw an expansion model of yellow peach moth populations, this study provide theoretical basis for further study on dispersal behavior population dynamics. The research of Wolbachia infection also lays a foundation for further study of reproductive manipulation mechanism between Wolbachia-host, providing a set of basic material to settle the issue about the relationship between Wolbachia infection and host differentiation.
