In this study, single nucleotide polymorphisms of MC4R/LPL/PPARy/ApoB genes in132geese from5species (Landes goose; Eastern Zhejiang white goose; Jiangshan white goose; Yongkang grey goose; Rhine goose) were detected by PCR-SSCP and DNA sequencing, and the effects of various genotypes on meat quality and carcass traits were studied by GLM analysis. In addition, the expression pattern of the MC4R gene was also investigated in overfeeding Landes geese, SYBR Green I real-time quantitative PCR was developed to analyze the expression characterization of MC4R gene in13tissues. The results were as follows:1. Study on the relationship between Polymorphisms of MC4R/LPL/ApoB Genes and Meat Quality/Carcass Traits.The results of PCR-SSCP showed that each one SNPs site in MC4R/LPL/ApoB gene was determined:SNPs-MC4R128(Gâ†'A), SNPs-LPLexton3-246(Gâ†'A), SNPs-apoB4507(Aâ†'G). There were2genotypes in SNPs-MC4R128(Gâ†'A) site (AA/BB) and each3genotypes in SNPs-LPLexton3-246(Gâ†'A)(HH/Hh/hh) and SNPs-apoB4507(Aâ†'G)(YY/Yy/yy). By the least squares means (LSM), there were significant differences between AA and BB in BMP and LMP (P<0.01). It had very significant difference between HH, Hh and hh in DP, LW and AFP (P<0.01), and, also had very significant difference between YY, Yy and yy genotype (P<0.01) in BMP, LMP, DP, LW and AFP.It is also showed that it had very significant (P<0.01) or significant(P<0.05) difference with some fatty acids between the genotypes in SNPs-MC4R128, SNPs-LPLexton3-246and SNPs-apoB4507. The present study proved that The MC4R, LPL and ApoB genes could be candidate modifier genes that affect or control the meat quality and carcass traits of goose.2. Expression pattern of MC4R gene by SYBR Green I real-time quantitative PCR in overfeeding Landes Geese.By PCR, it was showed that both MC4R and β-actin genes expressed in all13tissues before and after overfeeding in Landes Geese. By SYBR Green I real-time quantitative PCR, the results showed that the relative expression of MC4R in kidney, intestine and heart of overfeeding geese is lower than those of control,2"AACT values are0.41±1.80,0.65±5.75and0.72±1.22, respectively. But the relative expressions of MC4R in other tissues were upregulated after overfeeding,2-△△CT values of other tissues from high to low were: brain22.78±1.00, liver9.08±2.80, lung5.28±1.83,pancreas3.78±3.01,leg muscle3.07±3.64, stomach2.90±0.97, subcutaneous adipose2.34±1.66, abdominal adipose2.18±3.01, chest muscle2.07±0.37, pleen2.01±1.75,respectively.
