This study was conducted to investigate effect calcium pyruvate supplementation on fatty acid content, composition, lipometabolism, and ACC、SCD mRNA expression in liver of rats, on body weight and lipid metabolism in rats, to provide theoretical basis for its application in animal production.1Effects of Calcium Pyruvate on body weight and biochemical parameters in ratsThirty-two150g male SD rats were randomly divided into four groups:control group、 low、middle and high dosage group(0、100、500and1000mg/Kg. d-1). Calcium pyruvate was administered intragastrically to rats lasting for four weeks, and then rats were killed and blood and tissue samples were collected. Serum lipids and blood glucose of rats were determined by biochemistry analyzer. The results showed that at the end of the experiment, the body weight and food conversion efficiency of all experiment groups were significantly lower than that of control group. TG of high and middle dose group were obviously lower than that of control group. TC and blood glucose concentration were no statistical difference among different groups. Compared with control group, the three doses had no obvious change for different fat pad and fatty weight; for RFP in L, M, H groups decreased by16.03%,27.15%and34.68%, respectively; for EFP in L, M, H groups decreased by0.91%,11.01%,28.03%, respectively; for CFP in L, M, H groups decreased by11.91%,19.27%and26.97%, respectively. The ratio of fatty weight and body weight in L, M, H groups decreased by23.00%,7.52%and8.40%, respectively.2Effects of different dose of calcium pyruvate on fatty acid composition of different tissues in ratsThirty-two150g male SD rats were randomly divided into four groups:control group、 low、middle and high dosage group(0、100、500and1000mg/Kg.d-1). Calcium pyruvate was administered intragastrically to rats lasting for four weeks, and then rats were killed and blood and tissue samples were collected. Analysis of fatty acid composition of adipose tissue, liver tissue and musculature was by Capillary Gas Chromatography. Contents of C16:0, c9-C18:1, c9, cl2-C18:2, α-C18:3and C20:4in the adipose tissue were significantly decreased in the experiment groups, but the content of C18:0was no statistical difference. Fatty acid composition of liver tissue was remarkably lower than that of the control group, except contents of C18:0and cl2-C18:2. Effects of different dose of calcium pyruvate on fatty acid composition of musculature in rats was no statistical difference.3Gene expressions of ACC and SCD in liver of rats supplemented with Calcium pyruvateThirty-two150g male SD rats were randomly divided into four groups:control group、 low、middle and high dosage group (0、100、500and1000mg/Kg. d-1). Calcium pyruvate was administered intragastrically to rats lasting for four weeks, and then rats were killed and livers were collected. mRNA expressions of ACC-a and SCD in the liver was determined by RT-PCR with β-actin rRNA as internal standard. Compared with control group, ACC and SCD mRNA gene expression was significantly decreased in liver of high dose group (P<0.05). Additionally, gene expression of ACC was decreased by16.53%and9.45%in middle dose and low dose groups. SCD mRNA was decreased by18.25%and8.18%in in middle dose and low dose groups.
