The direct sequencing was applied to detect and compare the genetic diversity and structure amongLF,XX and JN populations of Misgurnus anguillicaudatus from the Yellow River basin.The fragments ofmitochondria cytochrome oxidase subunit I(COI)gene、12S and16S rRNA gene of Misgurnusanguillicaudatus from different areas were amplified by using PCR technique.The results were as belows:1. The mitochondria cytochrome oxidase submit(COI) fragments of Misgurnus anguillicaudatuscoming from LF,XX and JN were amplified via Polymerase Chain Reaction.The PCR products wereligated into T-vector,cloned and sequenced.668bp nucleotide sequences were gotten. Seventy-onepolymorphic sites and fifteen haplotypes were detected out of29individuals from these threepopulations.The average contents of T、C、A、G were32.5%、26.5%、22.8%and18.2%,separately.Thecontent of GC was higher than AT obviously.The haplotype diversity (H), the mean nucleotide diversity (Pi)and the average number of nucleotide differences (K) reached0.892,0.01464and9.749, respectively,which showed abundant genetic diversity among the three populations.The Kimura2-parameter averagegenetic distance of total population was1.6%calculated by the MEGA version4.0.The genetic distancewithin population ranged from0.3%to3.8%. The genetic distance among population ranged from0.4%to2.4%.The population of LF and the others was high,ranged from2.3%to3.8%.Fst values amongpopulation ranged from0.12303to0.14197,and the Nm was3.02187to3.56405.They means that thedifferentiated among population reach the level of moderately differentiated..AMOVA analysis(Fst=0.13146,P<0.01) revealed a strong genetic structure in these Misgurnus anguillicaudatuspopulations. The phylogenetic tree revealed and the median-joining network no obvious geographical pattern for the prevalence and distribution of the haplotypes.The genetic distance between Hap-11、Hap-13and other haplotypes reached0.096, indicating that they are different species. Fu’Fst and Tajima’s D testresult suggest that for the Misgurnus anguillicaudatus, there might be no population expansion and accordwith neutral mutations.2. A509bp long partial gene segment was acquired from the16S rRNA gene were amplified byPolymerase Chain Reaction.Sixteen polymorphic sites and sixteen haplotypes were detected out of30individuals from these three populations.The average contents of T、C、A、G were23.4%、22.9%、31.4%and22.3%,separately.The content of GC was higher than AT obviously.The haplotype diversity (H), themean nucleotide diversity (Pi) and the average number of nucleotide differences (K) reached0.906,0.00402and2.032, respectively.The Kirnura2-parameter genetic distance are less than2.0%calculated bythe MEGA version4.0.Fst values among population ranged from0.17942to0.38839,and the Nm was0.78737to2.28676.They and AMOVA analysis (Fst=0.28062,p<0.01)showed obvious geneticdifferentiation between the three populations.The phylogenetic tree revealed and the median-joiningnetwork no obvious geographical pattern for the prevalence and distribution of the haplotypes. Fu’Fst andTajima’s D test result Show that Misgurnus anguillicaudatus populations may have undergone populationexpansion events.3. A399bp long partial gene segment was acquired from the12S rRNA gene were amplified byPolymerase Chain Reaction. Seventeen polymorphic sites and thirteen haplotypes were detected out of34individuals from these three populations. The content of GC was higher than AT obviously.The haplotypediversity (H), the mean nucleotide diversity (Pi) and the average number of nucleotide differences (K)reached0.718,0.0032and1.273,respectively.The Kirnura2-parameter genetic distance are less than2.0% calculated by the MEGA version4.0.Fst values among population ranged from0.02093to0.16993,and theNm was2.44239to23.38915.They and AMOVA analysis (Fst=0.08027,p>0.05)showed indistinctivegenetic differentiation between the three populations.The phylogenetic tree revealed and themedian-joining network no obvious geographical pattern for the prevalence and distribution of thehaplotypes.Fu’Fst and Tajima’s D test result suggest that for the Misgurnus anguillicaudatus,there might beno population expansion and accord with neutral mutations.
