The transgenic strategy to improve P-use efficiency by crop plants has the potential toenhance sustainability of agricultural production and environmental management. We usedath-miR399d transgenic tomato (C1), grf9transgenic tomato (GRF9) and wild type tomato(WT) as the experiment plant materials, through soil culture and solution culture to explorethe potential of ath-miR399d and grf9gene could improve tomato growth and P-useefficiency, and study the physiological mechanism on how grf9gene improve tomato P-useefficiency. Some important conclusions are as follows:Firstly, in the150mg/kg P2O5treatment, leaf dry weight (LDW), stem dry weight (SDW)and shoot dry weight (SHDW) of ath-miR399d transgenic tomato were higher than wild typesignificantly, LDW, SDW, and SHDW of grf9transgenic tomato were higher than wild type,about10-20%; in the300mg/kg P2O5treatment, LDW, SDW and SHDW of ath-miR399dtransgenic tomato had no significant difference comparing to wild type, LDW and SHDW ofgrf9transgenic tomato compared to wild type reached up to20-30%.Secondly, in the150mg/kg P2O5treatment, shoot P content of ath-miR399d transgenicand grf9transgenic tomato were both higher than wild type significantly; in the300mg/kgP2O5treatment, shoot P content of ath-miR399d transgnic tomato had no significantdifference comparing to wild type, shoot P content of grf9transgenic tomato were higher thanwild type about56.68%.Thirdly, in the150mg/kg P2O5treatment, soil available P after plant ath-miR399dtransgenic tomato and grf9transgenic tomato both had no significant difference comparing towild type; in the300mg/kg P2O5treatment, soil available P after plant ath-miR399dtransgenic tomato were higher than wild type significantly and soil available P after plant grf9transgenic tomato were lower than wild type significantly.Fourthly, we could obtain from the solution culture that in the P200treatment, shoot dryweight and plant dry weight of grf9transgenic tomato had a increasing tendency comparing towild type, and increased up to30%; shoot P content and plant P content also had a increasingtendency, and reached to36-43%.Fifthly, we also could get from the solution culture, both in the P5and P200treatments,total root length, main root length and root surface area all higher than wild type, but the pHof solution were all lower than wild type. Sixthly, otherwise we found that as the soil P level increased, ath-miR399d transgenictomato produced the P intoxication but grf9transgenic tomato did not.
