In this paper, the cell suspension culture induced by Salvia miltiorrhiza Bung asepticleaves is used as the experimental materials. The present work was aiming at followingstudys: to observe the growth of S. miltiorrhiza suspension cell, synthesis of rosmarinic acidactivities of related enzymes (PAL and TAT) under the influence of salicylic acid andmethyl jasmonate, elicitors;to investigate the function of the signal moleculars (H2O2andMeJA) during the production of rosmarinic acid triggered by salicylic acid.The results lay a theoritical foundation on enhancement of production of the rosmarinicacid and provide evidence on investigating the rosmarinic acid accumulaton. And results areas follows:1. The datas showed that salicylic acid and methyl jasmonate could enhance theaccumulation of rosmarinic acid significantly. However, their optimum elicitation dosagevary greatly. The yields of rosmarinic acid significantly increased in the presence of lowconcentration,0.04525mmol/L SA on day6up to1.36-fold, whereas, in the presence of10μmol/L MeJA the maximum content of rosmarinic acid was2.743-fold than that of thecontrol group. Besides, the cultivation of S. miltiorrhiza cell suspension after elicitation withMeJA (10200μmol/L) may be valuable to enhance production of the rosmarinic acid. Andthe MeJA elicitor with high concentration (150200μmol/L) could suppress the cellsuspension cultures biomass accumulation, leaving the medium turbid.2. The datas revealed that, during the process of rosmarinic acid accumulation in thecell suspension cultures, elicitors (SA and MeJA) could change the enzyme (PAL and TAT)activity. Preceding the elicitor-induced rosmarinic acid accumulation, there was a notablerise in the PAL activity but slight rise in the TAT activity of suspension cells. Thus weconcluded the rate-limiting effect of PAL was stronger than that of TAT.3. The results revealed that, during the process of rosmarinic acid accumulation in the cell suspension cultures, SA, as the elicitor, could cause the cell of S. miltiorrhiza to producesignal molecular (H2O2and MeJA). And after treating the cell suspension cultures with IBU,a hibitor of MeJA, and exogenous catalase, a remover of H2O2, we found that both treatedgroups inhibited the rosmarinic acid accumulation in the cell suspension cultures. Andapplication of IBU can inhibit the signal molecular of H2O2.While, application of exogenouscatalase has no effect on production of MeJA. So we inferred that both of the signalmoleculars play significant roles in the signal transduction pathway of production rosmarinicacid. And MeJA maybe the upstream signal molecular of H2O2.
