Orphanin FQ (OFQ) is an endogenous ligand of opioid-like orphan receptor (0RL1), discovered in1995. Study shows that OFQ antibody can completely flip acute acupuncture tolerance,50%of the chronic acupuncture. This experiment studyed the produce, sustained and recovery process of acute acupuncture tolerance in goats. Assayed the OFQ concentration in nuclei and spinal cord, and then combined with the variation of pain threshold to conjecture the role of OFQ played in acute acupuncture tolerance.The experiments selected42the hybridization healthy goat, weighing30-35kg, were randomly divided into2groups, electro-acupuncture group and control group. EA group stimulate in the60Hz frequency electro-acupuncture, and this group contain EA stimulating0.5h,2h,4h,6h and stop EA stimulation12h,24h. Determinated of pain threshold and take the striatum, hypothalamus, septal area, the PAG and spinal cord. Determination concentration of OFQ by ELISA, and explore the variation of the concentration of OFQ in the EA process.The pain threshold test showed that:after EA stimulate0.5h, goats get the best analgesic effect, and then began to decrease, to2h after the pain threshold is reduced to below the pain threshold of control group, alone with EA stimulation continuous to4h, the pain threshold at the lowest level. After stop EA stimulation24h, the pain threshold recovered to the nomal level.Determination of OFQ concentration in the striatum, hypothalamus, septal area, PAG and the spinal cord, showed that OFQ in the PAG and striatum have been a substantial change of concentrations in EA process. Concentration of OFQ in Striatum have a slight reduced in the time of EA stimulation0.5h, after EA stimulation2h, there has been a great rise in the concentration than the control group. Concentration of OFQ in hypothalamus and septal area in the process of change is not significant as PAG. The concentration of OFQ in the spinal cord has no significantly increased except with EA stimulation2hours to the control group.Correlation analysis OFQ concentration and pain threshold use SPSS18.0statistical software, and found that the relationship between the release of endogenous OFQ in different nuclei in the solution of goat brain. Find that the concentration of OFQ in PAG has height correlation with pain threshold, with the spinal cord is low correlation and the other is midrange correlation.
