Ramie (Bochmeria nivea L.) is a perennial basic fiber crop of the Urticaceae Bochmeria.lt has a long history of cultivation and processing in our country and is known as’China grass’ in the world. Presently, the task of breeding is very difficult. The key problem is lack of good homozygous parent material. Luckily, the wild resources is very rich and not yet utilized by man. Therefore, it is very important to research the genetic diversity of wild resources, in order to explore the good genes related. Meanwhile, it is very significant to improve the level and efficiency of the breeding task of ramie. We have conducted a research in the wild genetic diversity of ramie. The results were reported as follows:1.We have been Yunnan、Sichuan、Jiangxi and Hubei province for many times to collect the wild materials. At present, we have collected about150materials(belong to9kinds)(see appendix1)and established the wild species resources nursery of ramie in HZAU successfully.2.We use FCM to determine the fluorescence intensity which the DAPI of rice(ZH11) and ramie sends up, comparing the peak-ploidy,then calculate the genome size. We repeat the test and calculate the genome size. The genome size of Urticaceae is about717.67Mb,or2C DNA about1.47pg(1pg DNA=0.978X109bp).The2C DNA of B. macrophylla is1.05pg. The2C DNA of B.nivea is1.47pg, B.longispica is1.41pg, B. tricuspis is1.32pg,B.huazhu NO.5is1.05pg.The result of the study is the first report about the genome size and will fill the information of genome of ramie. At the same time, it will provide the basic research of genome size of ramie.3.We analyze the wild materials of ramie(5kinds) in cpDNA, which can be proved that the amplification product is credible. The phylogenetic tree using the MEGE N-J method. The result were reported:the Urticaceae and the Ramie gather two groups, which can be proved that the cluster analysis is credible. In the0.007level, the materials of ramie gather two groups that divide into several subgroups. The B.nivea and the B. tricuspis gather together. The B. macrophylla and the B.longispica gather2,3subgroups.The B.huazhu NO.5and the wild materials were divided.
