The Taishan pine pollen, Taishan Robinia Pseudoacacia, Firmiana simplex flowers andAloe contain a number of essential elements of life and they are full of rich nutrients andactive substances. The Taishan pine pollen, Taishan Robinia Pseudoacacia, Firmiana simplexflowers and Aloe also own the function of antibacterial, antiviral, antioxidant,immune-enhancement, anti-tumor and anti-HIV which are great benefit to human health. Inrecent years, the development of Chinese animal husbandry has made a remarkableachievement. But the increasing of antibiotics is used for the economic efficiency and thisresults that drug residue. And the drug residue has seriously restricted the export of animalproducts of China. Therefore, recent years, researching and development of natural-plantimmunostimulants are becoming one of the directions of livestock and poultry industry. Inview of previous related research, this study focused on the important bacteria and viruses andstudies the effects of four polysaccharides on antibacterial, antiviral in order to provide theexperimental basis for the development of natural-green immunostimulants. Many studieshave demonstrated that a variety of traditional Chinese medicine polysaccharide have thefunction of immunostimulants. In our laboratory, Wei Kai and Sun Zhen Hong, who hasproved that the four kind of plant polysaccharide have the function of immuno-stimulantsNow, there are many reports of a certain Chinese medicines on the immunostimulants. Butthere are rarely reported about the comparing with the biological activities of immunizationand there are reported about the characteristics of polysaccharide. Therefore, our group usespropolis as a control to study and compare the respective immune characteristics of theTaishan pine pollen, Taishan Robinia Pseudoacacia, Firmiana simplex flowers and Aloe. Thisstudy provides experimental basis for the development of new immune enhancer and poultryvaccine adjuvants.This research consists of two parts as follows:Part One: Study on bacteriostasis and antivirus effects of four polysaccharidesBacteriostasis to pathogenic bacteria of four polysaccharides was studied by using thedrilled nutrient agar medium to observe the pathogenic bacteria of different polysaccharides; this test studied antivirus effect of the polysaccharides to IBDV and NDV using embryoinoculation methods. The results showed that: the polysaccharides of Taishan RobiniaPseudoacacia, Taishan pine pollen, Firmiana simplex flowers and Aloe have antibacterialeffects in this study. The bacteriostasis of Taishan Robinia Pseudoacacia Polysaccharide toStaphylococcus aureus, Sarcina, Escherichia coli and Bacillus cereus was inhibitory zonediameter longer than14mm; the bacteriostasis of Taishan pine pollen Polysaccharide toSalmonella pullorum was the best, inhibitory zone diameter longer than16mm, thebacteriostasis to Bacillus cereus, Proteus mirabilis, Bordetella avium was inhibitory zonediameter longer than14mm; The bacteriostasis of Firmiana simplex flowers Polysaccharide toBordetella avium, Sarcina, Bacterium coli, Tetrads was inhibitory zone diameter longer than16mm, The bacteriostasis to Bacillus cereus was inhibitory zone diameter longer than14mm;The bacteriostasis of Aloe Polysaccharide to Bacillus cereus was inhibitory zone diameterlonger than16mm, The bacteriostasis to Tetrads, Sarcina, Staphylococci aureus, Bacillusaeruginosus was inhibitory zone diameter longer than14mm. The HA results of experimentalgroups were lower than virus control group. With the increase of concentration and actiontime, HA results became lower. The concentration of Taishan Robinia Pseudoacaciapolysaccharide, Taishan pine pollen polysaccharide, Firmiana simplex flowers polysaccharideand aloe polysaccharide were5000mg/L and then were mixed with NDV2h. The HA resultsof Taishan Robinia Pseudoacacia polysaccharide, Taishan pine pollen polysaccharide and aloepolysaccharide reduces four units, the HA result of Firmiana simplex flowers polysaccharidereduce the three units.Part Two: Comparative study the effect of four kinds of Polysaccharides on immuneenhancement bordetella bronchiseptica in RabbitBordetella bronchiseptica was cultured by nutrient agar and then it was inactivated byformaldehyde. Propolis adjuvant and Taishan Robinia Pseudoacacia polysaccharide, Taishanpine pollen polysaccharide, Firmiana simplex flowers polysaccharide and aloe polysaccharideextracted by the modified water extraction and alcohol precipitation method were added toBordetella bronchiseptica.54rabbits were randomly divided into six groups, I, II, III, IV, wasrespectively immunization with Taishan Robinia Pseudoacacia polysaccharide, Taishan pine pollen polysaccharide, aloe polysaccharides, Firmiana simplex flowers polysaccharide rabbitBordetella bronchiseptica bacteria inactivated-vaccine; Ⅴ group was immunization withPropolis rabbit Bordetella bronchiseptica bacteria inactivated vaccine; VI group was justimmunization with rabbit bronchial Bordetella inactivated vaccine. The samples of bloodwere then taken from the rabbits3,7,14,21,28,35,42day after immunization. Serumantibody titers determined by indirect ELISA method, lymphocyte ratio, lymphocyte count byautomated hematology analyzer, blood lymphocyte transformation rate by flow cytometry. Weuse rabbit interleukin-2(IL-2) enzyme-linked immunosorbent assay kit to determine thecontent of interleukin-2(IL-2) in serum. The results showed that: generally, the effects of thepolysaccharide group were better than the propolis group and negative control group. Exceptthe content of serum interleukin2(IL-2),the immune-enhancing of Taishan pine pollenpolysaccharide was the best Firmiana simplex flowers polysaccharide, Taishan RobiniaPseudoacacia polysaccharide took the second place; the immune-enhancing of aloepolysaccharides, is worse than another three kinds of polysaccharides, but its effect is fargreater than the other three kinds of polysaccharide in serum IL-2content.
