To enhance the dairy goat genetic improvement and provide theory for production traits MAS of dairy goat,436dairy goats and48black goats as a research object in this study, PCR-SSCP was applied to analyze the genetic polymorphisms at exon9and exon14of ABCG2gene of milk goat; DNA mixed pool technology and High resolution melted curve analysis(HRM) technology were used to detect the genetic polymorphisms at intro5,exon6and exon7of OPN gene of lactating ewe; genetic polymorphisms of polymorphism sites were analyzed by Pop3.2software; the polymorphism information content(PIC) was calculated through Molkin3.0; correlation analysis between the mutation sites of Laoshan dairy goat and Wendeng dairy goat and the lactation trait and growth trait is analyzed by SAS9.2;346milk goat and48black goat were used in this study. The result showed:(1) the mutation site that relate to milk performance were not detected at exon9and exon14of ABCG2gene of milk goat which is different from dairy cow; one mutation site was detected at intron5and exon7separately, which is the C/T of locus1355located in intron5and M/N of lucus E341in exon7. The PIC of Laoshan dairy goat population are0.2773and0.3267respectively, which all belong to an intermediate polymorphism. The allele frequencies and genotype frequencies of two SNPs were in Hardy-Weinberg equilibrium on the distribution of the breeding farm (F) and breeding households (P) groups except the P group of locus1355(P<0.05).(2) The phenotypic value of all trait in P groups were extreme significantly (P<0.01) higher than the F group except conductance traits. The fat content trait of the CC genotype was significantly (P<0.05) higher than CT and TT genotype, while the ash and freezing traits significant (P<0.05) higher than the CT genotype at locus1355. Allele N had a positive effect on non-fat rate, lactose rate, milk density, milk protein percentage, the freezing point and ash, while all the traits at locus E341were not significantly different (P>0.05).(3) Analysis associated with growth traits of Laoshan Goats, the body weight and body length of the F group are extremely significant (P<0.01) higher than P group. Rump width and rump length of the P group was significantly (P<0.01) higher than the F group. The weight of the TT genotype extremely significant (P<0.01) greater than the CC genotype at locus1355. Allele M was the dominant allele for body weight and heart girth at locus E341. The weight and rump width of the MM and MN genotype extremely significant (P<0.01) higher than NN genotype. The body length of MM genotype was extreme significantly (P<0.01) higher than that of genotype NN, and the body height and chest circumference of MM genotype was significantly (P<0.05) higher than that of genotype NN. Analysis associated with growth traits of Wendeng Goats,1355is significantly (P<0.05) with the Primiparity goat’s rump width and is significantly (P<0.05) with Multiparity goat’s body height and body length;1355is significantly (P<0.05) with the Primiparity goat’s body length and is significantly (P<0.05) with Multiparity goat’s bust circumference.This study found that OPN gene is related to fat and carbohydrate content of milk, the OPN gene has significant effect on milk goat’s growth and development traits that can be used as molecular genetic markers for part of the production traits of milk goats.
