In this study, investigation and sampling was developed on tomato-growing region in Jiangsu province, samples were sequenced and analysised, The results indicated that the disease had occurred in Nanjing, Suzhou, Changzhou, Xuzhou, Taizhou and Yancheng. Sequence analysis revealed that the samples shared over99.0%nucleotide sequence identities with tomato yellow leaf curl virus(TYLCV).Xinghua virus isolates were cloned and sequenced, BLAST analysis revealed that the samples were tomato yellow leaf curl virus. There were no DNAPexcept DNA-A by molecular detection. In order to clear the infection of DNA-A of TYLCV in Jiangsu, DNA-A was infectious cloned, the construction was shifed to agrobacterium to injection tomatos and tabaccos, infectious verification showed that the virus of TYLCV was the pathogeny which caused tomato yellow leaf curl virus in Jiangsu province.9varieties were injected by agroinoculation of infectious clone to screen resistence varieties of TYLCV, at the same time, vector whitefly inoculation was made comparision. The result revealed that agroinoculation and whitefly inoculation had the same disease severity. And we attained two varieties of high resistence to TYLCV, which provided resistent parent materials for breeding.Put the construction of infectious clone into agrobacterium and agroinoculation tomatos to screed resistence was the fist report in China. To comparied with whitefly inoculation, agroinoculation could make consistent with ammout vaccination and period, the result was more accurate and stable. The application of agroinoculation provided a accurate and steady way to screen resistence tomato varities for TYLCV.
